Vitiligo is a relatively common, acquired pigmentary disorder characterized by areas of depigmented skin, The prevalence of the disease is 0.1%-2%, Without preference of gender. The pathogenesis of vitiligo remains unclear. There are four hypotheses, autoimmune, genetic neural and melanocyte self-destruction. Researches showed that, vitiligo always associate with autoimmune diseases which prompted studies on the association between HLA and vitiligo. Studies have showed that the association of HLA with vitiligo varies among different ethnic groups. The frequency of HLA-DRB 1*0701,1201 alleles have increased in mulititudinous researches. Under the background of Key research projects and Key technology project of Autonomous region, we did a preliminary research to detect these two alleles in vitiligo of Uyghur population in xinjiang region.Object:To detect HLA-DRB 1*0701,DRB1*1201 alleles in different onset age,with or without familiy history,clinical type,clinical phase, To identify the association with vitiligo in the Uyghur nationality of Xinjiang region.Methods:Extract genomic DNA, Use polymerase chain reaction sequence-specific primer (PCR-SSP) to detect HLA-DRB1*0701,DRB1*1201 of 300 vitiligo patients and 300 healthy person in the Uyghur nationality of Xinjiang region.Result:1,Compared with control group HLA-DRB1*0701,DRB1*1201 alleles has a high frequency, the difference was significant.2,HLA-DRB1* 0701 alleles increased in both childhood-onset (onset age〈12) case and adulthood-onset(onset age〉12) case compared with control group, the difference was significant; Frequency of HLA-DRB1*0701 alleles appears no difference between childhood-onset case and adulthood-onset case; HLA-DRB1*1201 alleles has a high frequency in both childhood-onset and adulthood-onset case, the difference was significant. HLA-DRB1*1201 alleles has a high frequency in childthood-onset case, compared with the adulthood-onset case.3,HLA-DRB1*0701 alleles has a high frequency in positive familiy history and negative family History cases, the difference was significant; HLA-DRB1*1201 alleles only has a high frequency in negative familiy history, there is no difference in positive familiy history compared with control group.4,HLA-DRB1*0701 alleles appear to be a high frequency in focal and generalized vitiligo, the difference was significant; HLA-DRB1*0701 alleles has a high frequency in generalized vitiligo compared with focal vitiligo; HLA-DRB1*1201 alleles has a high frequency in focal and generalized vitiligo, the difference was significant.5,HLA-DRB1*0701 alleles has a high frequency in progressive and stable phase, the difference was significant; HLA-DRB1*1201 has a high frequency in progressive phase, the difference was significant. There is no difference in stable phase compared with control group.Conclusion:HLA-DRB1*0701,1201 may be the susceptible alleles of vitiligo in Uyghur nationality of Xinjiang region.HLA-DRB1*0701 alleles may be the susceptible alleles of generalized vitiligo in Uyghur population. HLA-DRB1*1201 alleles may be the susceptible alleles in childhood-onset and progressive vitiligo.
