Objective To study the expressions of DSCR1gene in laryngopharynx squamous cell carcinoma tissues and adjacent tissues to the cancer, to understand the relationship between its expressions and tumor clinical features, to discusse the influence of DSCR1gene on the biological behaviour of laryngopharynx squamous cell carcinoma. To provide biological basis for DSCR1gene target antineoplastic therapy.Methods Collected94cases of laryngopharynx squamous cell carcinoma,78cases were also taken adjacent tissues, Immunohistochemical p-V9000method,used rabbit DSCR1antibody DCT3, detected the expressions of DSCR1gene protein in laryngopharynx squamous cell carcinoma tissues and adjacent tissues to the tumor. Data was inputed SAS8.2software for statistical analysis, a chi-square test was used to compare the expression differences between groups of gender, age, histological grade, the clinical stages, site, growth mode, lymph node metastasis and smoking history.Gene expression differences between tumors and adjacent tissues of the same78patients were analyzed by matching t test method., P<0.05for with a statistical significance Results Positive rate of DSCR1gene expressions in the tumor tissues was94.9%, normal tissues was35.9%, there was a significant statistically significant (t=23.69, P <0.001);There was significant correlation between DSCR1gene expression and pathological differentiation degree, TNM staging (P<0.05). There was no correlation between DSCR1gene expression and age, sex, site, growth mode, lymph node metastasis and smoking history (P>0.05)Conclusion1.There exist obvious differences of DSCR1gene protein expressions between the laryngopharynx tumor tissues and adjacent tissues to cancer.2.There are obvious correlation between DSCR1gene and tumor histological grade, The poorer histological grade degree, the higher the positive rate of DSCR1gene expression, the relationship of gene expression between TNM staging and lymph node metastasis need to be studied further.3.DSCR1gene has an important role in the occurrence and development process of laryngopharynx squamous cell carcinoma, may be cancer development is not controlled caused by the gene inactivation or dysfunction, and the gene affects tumor biology characteristic, is a promising cancer gene therapy targets. Objective By testing the microvascular density numerical of laryngopharynx cancer tissue and cancer adjacent tissues, to study the correlation between vascular factors and invasiveness of tumors. To make the correlation analysis with the earlier measured expressions of DSCR1gene, To discusse the relationship between DSCR1gene and the angiogenesis of laryngopharynx cancer,to study the influence by DSCR1gene on the happening, angiogenesis and other biological behaviour characteristics of laryngopharynx cancer. Methods Used immunohistochemistry method of PV9000, with the first part of the same94patients with laryngostenosis pharynx squamous cell carcinoma, through CD34antibody stained and markered Vascular endothelial cells, calculated the microvascular density (MVD) value, discusseed the relationship between MVD and the earlier measured expressions of DSCR1gene marked by DCT3antibody through the statistical analysis,MVD results were recorded by mean±standard deviation, between two groups, compared by the t-test, above three groups sample, used single factor analysis of variance, P<0.05for with a statistical significance.The Spearman correlation analysis was used in testing the correlation between DSCR1expression and MVD.Results Average numerical of MVD of laryngopharynx cancer tissue for swallow was17.57+10.61bar/400times each view, it was12.61±7.67/400times each view in tissues adjacent to cancer, there was significant difference by statistical analysis (P <0.05). It was18.96+11.63/400times each view in group of lymph node metastasis, however it was16.17+9.61/400times each view in group of without lymph node metastasis, there was significant difference by statistical analysis (P<0.05).It was16.29±9.98/400times each view in group of high pathology differentiation, and it was18.58±10.35/400times each view in group of medium pathology differentiation,while the MVD Valueis20.48±13.6/400times each view in group of low differentiation,there was significant difference by statistical analysis (P <0.05).The relevance research of blood vessel density MVD and the tumor clinical stages indicated that, MVD was16.13±9.61/400times each view in group of stages of Ⅰ~Ⅱ, but it was18.43±11.61/400times each view in group of stages of Ⅲ~Ⅳ, there was significant difference by statistical analysis (P<0.05).There was no correlation between MVD and age, sex, site, growth mode and smoking history (P>0.05).In the laryngopharynx squamous cell carcinoma tissues,along with the MVD ascension, the DSCR1gene expression strengthened gradually also, there was significant difference by statistical analysis (P<0.0001), and it was positive correlation between them(r=0.698). Conclusion1. In the laryngopharynx squamous cell carcinoma tissues MVD is concerned with tumor histological grade, TNM staging, lymph node metastasis. It explains that the different biological characters concomitance with different expression of tumor MVD, we can judge the prognosis of patients trouble with laryngopharynx squamous cell carcinoma tissues through testing the MVD, and it is necessary for patients with high MVD to sweep clear off cervical lymphatic glands for preventive,and postoperative radiotherapy and chemotherapy synthetic therapy are necessary too.2.In the laryngopharynx squamous cell carcinoma tissues, as the higher activity of DSCR1gene expression,the tumor microvascular density is reinforced, there exists positive correlation relationship, confirms the close correlation of DSCR1gene and tumor angiogenesis, explains that DSCR1gene can influence the process of invasion and transfer in laryngopharynx squamous cell carcinom, and it is a very promising therapeutic targets for cancer. Objective To realize the expressions of DSCR1mRNA and VEGF-CmRNA in the laryngopharynx squamous cell carcinoma and the normal tissues adjacent to cancer. Further to validate the relationship between VEGF and DSCR1gene in the tumor. And to make the correlation analysis between VEGF-CmRNA expression level and DSCR1mRNA level, to discusse the relationship between the DSCR1gene and the angiogenesis of laryngopharynx squamous cell carcinoma.Methods Detected the expressions of DSCR1mRNA and VEGF-CmRNA through the RT-PCR method in43patients with laryngopharynx squamous cell carcinoma and21cases of normal tissues adjacent to cancer. And compared the expressions between different histological grade, the clinical stages, lymph node metastasis, made the correlation analysis between them.Results35cases could be detected DSCR1-mRNA positive in43cases of the cancer, accounted for81.39%,8cases could be detected DSCR1-mRNA positive in21cases normal cancer adjacent tissues, accounted for38.10%. The average relative light density values of DSCR1-mRNA expression in tumor tissues was0.91±0.10, significantly higher than0.61+0.08of normal tissues of r adjacent to cance (P<0.05). There were significant differences of DSCR1-mRNA expressions in different histological grade, and lymph node metastasis.32cases could be detected VEGF-CmRNA positive in43cases of the cancer, accounted for74.42%,6cases could be detected DSCR1-mRNA positive in21cases normal tissues adjacent to cancer, accounted for28.57%. The average relative light density values of DSCR1-mRNA expression in tumor tissues was1.21±0.41, significantly higher than0.56±0.19of normal tissues adjacent to cancer (P<0.05). The correlation of expressions of DSCR1-mRNA and VEGF-CmRNA in43tumor tissues, was a positive correlation. after spearman the correlation analysis.Conclusion1.The expressions of the DSCR1mRNA are significant different in the laryngopharynx squamous cell carcinoma and adjacent tissues to the cancer. There are significant differences of DSCR1-mRNA expression in different histological grade and lymph node metastasis, this gene and biological actions of tumor are closely related.2.From adjacent tissues to tumor tissues, VEGF-CmRNA relative gray value is higher trend from, is significantly different. Explaining that there is a close relationship between VEGF-C and the occurrence and development of cancer.3.The expressions between DSCR1-mRNA and VEGF-CmRNA are positively associated in the laryngopharynx squamous cell carcinoma, there is a close relationship between DSCR1gene and angiogenesis of laryngopharynx squamous cell carcinoma.
