Objective:To study the infected situation of human herpesvirus 8 in cutaneous squamous cell carcinoma and esophageal squamous cellcarcinoma, and compare differences of HHV-8 between cutaneous squamous cell carcinoma and esophageal squamous cellcarcinoma, identify the infection of HHV-8 K1 gene subtypes and K15 allele subtypes. And to investigate the relationship between the infection of human herpesvirus 8 K1, K15 and squamous cell carcinoma preliminary.Methods:40 cases of skin squamous cell carcinoma,40 cases of esophageal squamous cell carcinoma paraffin-embedded tissues were HHV-8 DNA extraction, amplification, direct sequencing, using DNASTAR software, Crustal W and PH YLIP software packages on the sequencing results of phylogenetic analysis, to determine the HHV-8 K1 and K15 isoforms, and finally usingχ2 test and Fisher exact test results were statistically analyzed.Results:(K1 test results):①40 cases of skin squamous cell carcinoma in 9 cases (22.5%) infected with HHV-8 K1,40 patients with esophageal squamous cell carcinoma in 8 cases (20%) infected with HHV-8 Kl, HHV-8 in esophageal squamous cell carcinoma of the skin infection rates between the two groups no significant difference (P>0.05)②HHV-8 K1 subtype A in 1 case,8 cases of subtype C in esophageal squamous cell infected with HHV-8 K1 subtype A subtype 2 cases,5 cases of subtype C, other subtypes not found.(K15 test results):①. HHV-8 K15 allele was detected in 9(22.5%)of 40 cases cutaneous squamous cell carcinoma and 8 (20%) of 40 cases Esophagea Squamous cell carcinoma,②There was no significant difference for infecting HHV-8 between cutaneous squamous cell carcinoma and esophageal squamous cellcarcinoma (P>0.05).HHV-8 K15M was not be detected in our study.Conclusion:①HV-8 infects squamous cell carcinoma maybe is a random infection, but not direct reason for the development of squamous cell carcinoma.②But there was no significant difference for infecting HHV-8 between cutaneous squamous cell carcinoma and esophageal squamous cell carcinoma (P>0.05).③Squamous cell carcinoma subtype of HHV-8 infection is mainly K1-C subtypes and K15 P allele, there is part of the Kl-A subtypes.
