Objective:Oral cancer,one of common tumor,is difficult to cure,high-frequent to recur,and poor for prognosis.Early prevention,diagnosis and treatment are the effective ways to reduce the incidence rate of oral cancer and improve the survival rate and the prognosis of patients.Research shows that part of the oral cancers results from the oral precancerous lesions.The in-depth study of oral precancerous lesions,in particular,defining the influencing factors of the cancer,is of great guiding significance to the clinical practice.Epidermal growth factor receptor is expression product of proto-oncogene c-erbB-1,and also one of the 4 members in the receptor family.Epidermal growth factor receptor is a kind of catalytic receptor and the transmembrane cell surface sensor with tyrosine kinase activity.Being activated,epidermal growth factor receptor can initiate a series of cascade reaction conducted by intracellular signals,thus finally activate related nuclear protein,and promote the transition of normal cell cycle.Some research shows that epidermal growth factor receptor,in the process of tumor development such as angiogenesis,cell proliferation,and in the transfer,adhesion,apoptosis and inhibition of tumor cells,has significant importance.High expression of EGFR in cancer cells often marks further development or poor prognosis of the disease.Vascular endothelial growth factor,the mitogenic factor for endothelial cells specialty with the direct effects on vascular endothelial cells,is an important factor of raising angiogenesis.VEGF is a selective mitogen and Inducts the proliferation of endothelial cells and promotes the angiogenesis through union with the receptors of VEGF.And the angiogenesis is the essential condition of tumor growth and metastasis and has close relations with the occurrence,development and metastasis of tumor.This study is to detect the expression level of EGFR mRNA and VEGF mRNA in the tissue of oral lichen planus and oral squamous cell carcinoma and the normal tissue,analyze their relationship with the occurrence and development of oral cancer,and explore their roles in the occurrence,development and metastasis of oral cancer through the technology of reverse transcription-polymerase chain reaction.Methods:13 cases of carcinoma tissue of patients with oral squamous cell cancer,12 cases of mucosal tissue of patients with oral lichen planus,and 10 cases of normal tissue were extracted respectively,and quickly frozen in refrigerator with temperature of-70?.The technology of reverse transcription polymerase chain reaction was used to detect the expression level of EGFR mRNA and VEGF mRNA in carcinoma tissue and normal tissue of OSCC patients and that in the tissue of OLP.GAPDH was considered as reference of standards.Statistical analysis was done through the statistic software SPSS 19.0.P<0.05 showed significant difference in statistics.Results:1 the expression level of EGFR mRNA in the tissue of Oral squamous carcinoma was higher than that in the oral cavity compressed moss tissue and normal tissue.(P<0.05)?2.The expression level of VEGF mRNA in oral squamous carcinoma tissue was higher than that in oral cavity compressed moss tissue and normal tissue.(P<0.05).Conclusion:1.Higher expression level of EGFR mRNA in oral squamous cell carcinoma tissue than that in oral compressed moss tissue and normal tissue hints the possible relations between high expression of EGFR results from weakening of inhibition signal of EGFR and the occurrence,development and metastasis of oral cancer;In the process of Epithelial hyperplasia,EGFR signaling system is interfered,thus causing abnormal transduction of transmembrane signal,making the cells to grow out of control and worse,forming he tumor cells,and inhibiting the apoptosis. Its positive expression has certain reference to judge the metastasis and prognosis of the oral cancer.The excessive expression of EGFR mRNA has a certain role in the occurrence and development of oral squamous cell carcinomas.2.Higher expression level of VEGR mRNA in oral squamous cell carcinoma than that in oral cavity compressed moss tissue and normal tissue shows that VEGF can promote angiogenesis of oral mucosal squamous cell carcinomas,and then the growth,invasion and metastasis of oral mucosal squamous carcinoma.This hints VEGF may play a very important role in the occurrence,invasion and metastasis of oral cancer.Vascular endothelial growth factor may be a potential molecular target in the treatment of malignant tumor.
