ObjectiveTo determine the antioxidant capacity and contents of flavonoids of commonly consumed fruits, and to investigate the effects of fruit juices with different antioxidant capacity on the function of antioxidant system in the aged. Some components in pomegranate juice were separated and characterized.Methods1. The ferric reducing/antioxidant power assay was used to measure the antioxidant capacity of fruits.2. An HPLC procedure was optimized for determination of five flavonoids (including quercetin, luteolin, kaempferol, myricetin and apigenin) contained in fruits.2. Several fruits with different antioxidant capacity were selected for gavage experiment in rats, in which the dynamic changes of serum antioxidant capacity were determined after gavage.3. Aged Wistar rats and human volunteers were administrated orally with pomegranate juice or apple juice respectively daily. At the end of experiment, the function of antioxidant system was assessed.4. The pomegranate juice was extracted with ethyl acetate or n-BuOH for component analysis. HPLC-MS-MS technique was applied to characterize the possible structure of some components contained in the extracts.Results1.
