Study On The Effect Of Tomato Juice And/or Grape Juice On The Proliferation Of Human Prostate Carcinoma PC-3 Cell

Many epidemiologic observations suggest vegetables and fruits are consistently associated with lower risks of cancers at almost every site, which was related to the phytochemicals provided by them.Tomato and tomato juice has many phytochemicals such as lycopene, and grape and grape juice also has many phytochemicals such as resveratrol .There are many studies about the anti-tumor effect of lycopene and resveratrol of which the anti- prostate cancer is the most. But there have not the reports related to the effect of the tomato juice and grape juice on the human prostate carcinoma and the comparied effect with lycopene and resveratrol respectively. In this paper, we selected the human prostate carcinoma PC-3 cell as the subject and studied the effect of tomato juice and/or grape juice on the proliferation of PC-3 cell, and compared the effect of tomato juice and lycopene, the effect of grape juice and resveratrol, the effect of tomato juice and grape juice. The main results been completed are as follows:The experiment was devided six groups:the standard control group,the vehicle control group, three dose level groups and positive control group.The dose level groups were treated with tomato juice with different concentrations 40, 80, 120μl/ml for 48h and the content of lycopene was 2,4, 6μg/ml respectively.The positive control group was treated with lycopene(10.72μg/ml).The results show that the tomato juice and lycopene could markedly inhibit the growth of PC-3. They could induce the breakage of DNA strand of PC-3 cell and TUNEL positive cells were detectable.We also could detect apoptotic peaks by flow cytometry in experimental groups and lycopene group.The percentages of apoptosis cells were 2.8%, 12.7%, 27.2%, 13.2% respectively. Inducing apoptosis in PC-3 cell is the important anti- prostate cancer mechanism of the tomato juice and lycopene. The ratio of apoptosis relative gene Bcl-2/Bax is 1.15, 0.60, 0.27, 0.58 respectively. It revealed that the tomato juice and lycopene perhaps through modulating the expression of Bcl-2 and Bax to induce apoptosis.The effect be measured in experiments speaking above was enhanced with theincreasing of tomato juice, and the best effect of tomato juice is the high dose group(120 P l/rm\6 v g/ml). The effect of low dose group is weaker than that of the lycopene group while the content of lycopene is four times fewer than that of the positive group.There are no difference between the middle dose group and the lycopene group, while the content of lycopene is half times fewer than that of the positive group. This study revealed the anti-prostate cancer effect of tomato juice is not only related to lycopene but other phytochemicals which have the synergistical interection with lycopene.The experiment was devided six groups:the standard control group,the vehicle control group, three dose level groups and positive control group.The dose level groups were treated with grape juice with different concentrations 16 > 32 s 64 u I/ml for 48h and the content of resveratrol was 0.128^ 0.256 > 0.512 u g/ml respectively.The positive control group was treated with resveratrol (4.56 u g/ml).The results show that the grape juice and resveratrol could markedly inhibit the growth of PC-3.They could induce the breakage of DNA strand of PC-3 cell and TUNEL positive cells were detectable. We also could detect apoptotic peaks by flow cytometry in experimental groups and resvertrol group.The percentages of apoptosis cells were 6.0%, 18.7%, 30.0% > 12.7% respectively. Inducing apoptosis in PC-3 cell is the important anti- prostate cancer mechanism of the grape juice and resveratrol. The ratio of apoptosis relative gene Bcl-2/Bax is 1.06 n 0.50 ^ 0.19^ 1.02 respectively. It revealed that the grape juice and resveratrol perhaps through modulating the expression of Bcl-2 and Bax to induce apoptosis.The effect be measured in experiments speaking above was enhanced with the increasing of grape juice, and the best effect of grape juice is the high dose group(64 u 1/mk 0.512 u g/ml). The effect of low dose group is weaker than that of the resveratrol group while the content of resveratrol is thirty times fewer than that of the positive group. The effect of middle dose group is stronger than that of the resveratrol group while the content of resveratrol is fifteen times fewer than that of the positive group.This study revealed the anti-prostate cancer effect of tomato juice is not only related to lycopene but other phytochemicals which have the synergistical interection with lycopene.We have further studied the effect of tomato juice combined with grape juice on the proliferation of PC-3 cell and its possible mechanism.We combined the low dose of tomatojuice with middle dose of grape juice. The results show the inhibitive effect of combined group is higher than that of any group of tomato juice and grape juice.and it could induce the DNA damage of PC-3 markedly. The study show that tomato juice combined with grape juice could lead to DNA damage which was the possible mechanisms of inhibiting the growth of PC-3 cell.and the phytochemicals of the combined juice have synergistical interection each other.
