BackgroundPomegranate is the king of fruit which possesses edible value and medical value. It is discovered that there are a lot of flavonoids which possesses anti-oxidative effect in pomegranate. Concentration of anti-oxidative constituents in wine which is brewed from pomegranate is higher than in port wine. Research on anti-oxidative effect of pomegranate has been developed both at home and abroad in recent years. It is discovered that pomegranate juice or extraction of pomegranate juice can remove hydroxy radical and superoxide anion free radical, suppress LDL and lipid peroxidation, suppress the formation of artherosclerosis.ObjectivesTo study the content of effective anti-oxidative ingredients in pomegranate juice, anti-oxidative effect in vitro and the influence on the level of anti-oxidation of aging mice of pomegranate juice.Methods1. The determination of anti-oxidative ingredients in pomegranate juice: determine the content of antiscorbic acid in pomegranate juice with 2, 4-dinitro-phenylhydrazine method. Determine the content of total polyphenol in pomegranate juice with spectrophotometric method, using gallic acid as the standard substance. Determine the content of anthoxanthin in pomegranate juice with spectrophotometric method and rutin as the standard substance.2. The determination of anti-oxidative effect in vitro: Adopt total anti-oxidation capability(T-AOC), capability of deoxidizing, capability of removing DPPH free radical, capability of removing hydroxy radical, capability of removing superoxide anion free radical and the anti-oxidative capability in linoleic acid lipid overoxidation system as the indexes and choose alpha tocopherol and grape polyphenol which are proved to have strong anti-oxidative capability as the standard substance for contrast in vitro study.3. The determination of the influence on the level of anti-oxidation of aging mice of pomegranate juice: 72 aging mice, half female and half male, were randomly put in three groups: high dose group, low dose group and control group. Pomegranate juice or water were given orally every day at the following doses of 20ml/kg pomegranate juice,10ml/kg pomegranate juice and 20ml/kg distilled water. Obtain 6 mice from each group, pull out the eyeballs and execute the mice with luxation respectively at the end of the first week, the second week, the third week and the fourth week. Separare the blood serum after 3500r/min centrifugalization for 15 minutes, conserve the blood serum under refrigeration for the determination of total anti-oxidation capability(T-AOC), superoxide dismutase(SOD),glutathione peroxidase(GSH-PX) and malonaldehyde(MDA).Obtain livers and freeze them for the determination of T-AOC, SOD, GSH-PX and MDA.Results1. The content of antiscorbic acid in pomegranate juice is 43.8±1.5mg/g.The content of total polyphenol is 1545.2±17.7mg/L. The content of total flavone is 159.1±3.0mg/L.2. The total anti-oxidation capability(T-AOC), capability of deoxidizing, capability of removing DPPH free radical, capability of removing hydroxy radical, capability of removing superoxide anion free radical and the anti-oxidative capability in linoleic acid lipid overoxidation system of pomegranate juice are stronger than alpha tocopherol and grape polyphenol in vitro study.3. Compared with control group, the disparity between T-AOC of high dose group and T-AOC of control group had statistical significance. The disparity between the two began to have statistical significance from the third week. T-AOC, SOD and GSH-PX of blood serum and liver of high dose group and low dose group increased remarkably. The content of MDA of blood serum and liver degraded with no significant effect. There was a more noticeable effect on the level of anti-oxidation of aging mice of pomegranate juice in high dose group than in low dose group.Conclusions1. Pomegranate juice has strong total anti-oxidation capability(T-AOC), capability of deoxidizing, capability of removing DPPH free radical, hydroxy radical and superoxide anion free radical and the capability of suppressing lipid overoxidation. It is detected that the content of total polyphenol in pomegranate juice is 1545.2mg/L. The polyphenol in pomegranate juice maybe is the important functional ingredient. But it is also possible that several anti-oxidative ingredients play important parts through additional or cooperative effect.2. Pomegranate juice has strong influence on the level of anti-oxidation in aging mice. With the intake of pomegranate juice, the anti-oxidative capability of aging mice was elevated gradually.
