| The experiment synthesized the experience of the treatment in curing women's sterility and the report of inducing estrus in swine, cattle and sheep in veterinary clinics, twelve kinds of Chinese herbal medicines were screened out. On the basis of the traditional theory of native land pharmacology, the rational compatibility of medicines and the relation of medicine and animal reproductive endocrine, were observed and studied.Also the advantage of the modern scientific means were taken to observe and study the components of Chinese herbal medicine in prescription, rational composed herbal medicine, which made the medicines produce synergistic action.Compose eight recipes and apply to the small white rats, the total weight of the rat's uterus and ovary, middle and external diameter thickness of the uterine horn, the length of uterine horn and the histology changes of the ovary and uterus were surveyed. According to the influences of the different recipes in the small white rat's above-mentioned supervise index, the composed Chinese herbal medicines which had greater acceleration to the small white rat's reproductive systems were screened out. Take the composed herbal medicines and apply to the heathy jingba canines which were in the stage of anoestrus.Four millilitres decocted Chinese herbal medicine were fed to each canine through out twenty days.The changes of canine's behavior appearance and vagina lymph were observed daily. During the treatment with the medicine vein blood samples were taken and the concentration of estradiol, progesterone and IL-1, IL-6 were measured. The experimental dogs were killed after days 14, 16, 20and 25 of treatment. their ovaries, uterus and vaginas were collected.and made into the sections. The sections of these samples were examinated with optical microscope. The composed herbal medicines were made into injection and applid to induce estrus of canine. Meanwhile, the stability, security, quality standard and the pharmacokinetics of the injection were measured.The results for the induction of estrus by the medicines are as follows: 1.After treatments with different Chinese herbal medicines, the total weight of the uterus and ovary, the middle and external diameter of uterine horn and the proportion of big to middle follicles and the diameter of the biggest follicles all changed significantly which compared to the control group The results from the Chinese herbal medicine â…¡, â…£ and â…¤ were remarkably, different to the control group (p<0.01).The Chinese herbal medicine â…¡, â…£especially for the â…¤ had a significant accelerating function to the development of small white rat's reproductive system.2.Therefore, the Chinese herbal medicine â…¤ was selected to be the best prescription. The Chinese herbal prescription V induced female canine estrus, at the beginning of the estrus, E2 was 10pg/ml on an average, 5 days later E2 began to increase and at the 9 days it increased much quickly until the 15days.(It meant till 1 to 2 days before the end of estrus),it reached the peak level of 58.33pg/ml. then it declined quickly, until by the day of 25 reached to 18.17pg/ml.The concentration of P4 before taking medicine and in the early days before estrus were both low, 5 days later, the concentration was 0.12ng/ml, by the day 21 its concentration increased to 2.21ng/ml, afterwards it increased gradually and increased to 6.42ng/ml on the day of 25.3.After five days of treatment by the compound Chinese herbal medicineV, the concentration of IL-ip achieved to 0.578 ng/ml.and achived continuously, it achieved to the highest level of 0.830ng/ml by the day 21 around ovulation.lt declined to 0.608 ng/ml till 17 days.And yet at the five days of treatment, the concentration of IL-6 achieved to 0.163ng/ml, later it increased continuously to 0.312ng/ml on the day of 17.And its concentration reached to the highest of 0.348ng/ml by the day 21 around ovulation.But later it began to decline, it declined to 0.285ng/ml till 25 days.4.After several days of treatment by the compound Chinese herbal medicine V , fifty percent canines were observed vaginal bleeding. Red blood cells were present in large numbers of the vaginal bleeding dogs and its color changed to red and its concentration decreased. After using the Chinese herbal medicine 14 days, the thickness of vaginal epithelium, the weight of left, right ovary changed significantly (p<0.01), which compared to the control group; the circumferences of the left and right uterine horn and uterine body changed significantly(p<0.05); uterine slice revealed that uterine epithelium became edema.Several uterine glands in different size on the surface of the uterus were in different shapes.There were red cells in some glands. Follicular began to develop in the ovarian slice, and appeared follicle cavity. After seventeen days, the cell increased in the vaginal slice.The cell nuclear became blurred.Uterine slice revealed that endometrlUm became thicker and the grand proliferated. Ovarian slice appeared and the follicular cavity, follicle increased and formed granulose layer and ovarian mound..After 20 days, the cells keratinise completely in vaginal slice.The cell nuclear decreased in size.Uterine slice revealed that endometrlUm increased in thickness and became concave. The section of the surface grand appeared circle, the grand in depth became bigger, arranged together. It revealed the grand cell were in the stage of dynamic.Ovarian slice appeared mature ovulating follicle, the volume of ovary increased rapidly.The follicular cavity was so big that the diameter was up to 3-4mm. here were full of follicular liquid in follicular cavity. Follicie cell separated from ovarian mound, granulose layer became thinner, the follicle membrane was clearly seen.The follicle had come to the surface of the ovary and on the point of ovulation.5.The Chinese herbal prescription V were stable against light and heat. It didn't appear to be allergy against the skin of cavy. It had gentle stimulation to the eyes of rabbits. It was safe on the acute toxicity experiments giving orally or injected abdominally.LDso = 1920.84+331.66mg/kg, the 110 times of the biggest tolerance. It indicated that theChinese herbal prescription V injection was safe and reliable.6. The quality standard of experiment of Chinese herbal prescription V injection was executed by three different chromatograms.The injection contained tusizi, honghua, yimucao, yinyanghuo, nvzhenzi and xiangfu.Three different chromatograms can determine the injection.According to the ultraviolet absorption curve, and had a alkaloid compound absorption perk at 265-280nm wavelength, the wavelength absorption curve was also the characteristic absorption curve of the injection.7.The results from the pharmacokinetics experiment of Chinese herbal prescription V indicated that the Ka was 3.6769g/h or 2.6809g/h and the Tp was 1.1732h or 1.239h.Cmax was 3324.277ug/ml or 254.277ug/ml. kel was 0.0523/h or 0.111/h, it revealed that after the injection, the metabolize in rat had a character of quick absorption, and slow elimination.8.The Chinese herbal prescriptionVtaking orally and uterus pouring group, their estrus ratio were 50% and 58%, comparing to hormone selection of prescription A(75%) and B(83%)and the combination group of hormone and Chinese herbal medicine (75%), they were low, and the pregnant ration was 41% and 50%, which were all exceeded to A and B. The becoming pregnant ratio of hormone Chinese herbal medicine was 58%.It exceeded to orally taking group, uterus pouring, A and B group. |
