Bao Yuan Detoxofication Decoction Intervention Role On The Life Quality Of Cancer Cachexia Model And Affecting Atrogin-1, MuRF-1Expression

Objects：Copy the cancer cachexia model，evaluating their life-quality, to explorethe intervention effect of Bao Yuan detoxofication decoction in improving the qualityof life of cancer cachexia model animal. Detect the expression of Atrogin-l andMuRF-1which related with muscle atrophy, to explore Bao Yuan detoxoficationdecoction affecting the expression of Atrogin-l and MuRF-1.Methods：Establishing mice model of cancer cachexia by inoculating lewis cancer cellto the right axilla of C57BL/6mice subcutaneously.50mice were randomly dividedinto5groups: normal control group(A),model control group(B), TCM group(C),TCMprevention intervention group(D)and MPA group(E). Observing The changes of bodyweight,food intake,water intake,tumor volumes,tumor weight,gastrocnemius muscleweight and survival time.Observing Muscle fiber morphology byHE(hematoxylin-eosin staining); Detecting the3-methyl histidine level ingastrocnemius by HPLC(high performance liquid chromatography) with fluorescencedetection; Detecting the expression of Atrogin-1, MuRF-1mRNA and protein ingastrocnemius by HTC(immunohistochemical method), RT-PCR and Westernblot.Results：After drug treatment, compared with the B group, C group and D group,mice body weight, food intake and water intake are significantly increased; tumorvolume and tumor weight are significantly reduced; the gastrocnemius muscle weightand gastrocnemius muscle fiber diameter are significantly increased;3-MH aresignificantly decreased; the survival time are significantly prolonged; the expressionof Atrogin-1, MuRF-1mRNA and protein are significantly reduced (P<0.05). Compared with E group, C group and D group mice body weight, food intake andwater intake are increased (P>0.05); tumor volume and tumor weight are significantlyreduced; the gastrocnemius muscle weight and muscle fiber morphology aresignificantly increased;3-MH are significantly decreased; the survival time aresignificantly prolonged; the expression of Atrogin-1, MuRF-1mRNA and protein aresignificantly reduced (P<0.05).Conclusions：Bao Yuan detoxofication decoction is helpful to improve the quality oflife of cancer cachexia mice and reduces3-MH content and expression of Atrogin-1,MuRF-1in the gastrocnemius muscles of mice. Bao Yuan detoxofication decoctionimproves cachexia muscular atrophy status, prolonging life span.
