Objective:To study the early muscle changes after acute stroke and measure the Atrogin-1and MuRF-1mRNA level. Rats were used to make stroke model by middle occlusion.Method:60male wistar rats,10for the sham operation group (A), and the remaining50Longa suture method using rat middle cerebral artery occlusion (MCAO), of which30successful model rats were randomly divided into B, C, D groups of10. Group B was1day after successful modeling group, C group was4days after successful modeling group, D group was7days after successful modeling group. Application Bederson score evaluation of animal recovery after nerve injury; respectively for A, B, C, D four groups. each group of muscle fibers cross-sectional area were compared through HE staining; fluorescence quantitative RT-PCR was used to observe rat skeletal muscle atrogin-1and MuRF-1mRNA expression changes.Result:B, C, D rats Bederson score was significantly higher compared with group A (p<0.05); B, C, D group were no significant differences (p>0.05). Paralyzed muscle HE staining showed:A, B, C three groups of muscle fiber cross-sectional area pairwise comparisons, the difference was not statistically significant (p>0.05); D rats muscle fiber cross-sectional area was compared with the other three groups, the differences were statistically significant (p<0.05). B, C, D group Atrogin-1and MuRF-1mRNA expression levels were compared with control group, the differences were statistically significant (p<0.05); B group and C group were compared with no significant difference (p>0.05); D rats, were compared respectively with B, C group, the differences were statistically significant (p<0.05).CONCLUSION:Cerebral ischemia-reperfusion injury in rat skeletal muscle morphology of early Paralyzed muscle has changed. Atrogin-1and MuRF-1mRNA have been expressed in cerebral ischemia-reperfusion injury rat skeletal muscle.early cerebral ischemia-reperfusion injury rat skeletal muscle ubiquitin ligase pathway of protein degradation may have been activated. |
