Objective:To observe morphological changes of myoblast through Bao yuan detoxoficationdecoction medicated serum intervened malnutrition model of skeletal muscle cell by TNF-α.Detecting the effect of Bao yuan detoxofication decoction medicated serum on expressionof Ubiquitin、E214k、Atrogin-1、MuRF-1mRNAand protein.Discussing the mechanism ofskeletal muscle protein degradation by Bao yuan detoxofication decoction medicatedserum.Methods:Medicated serum was prepared and analyzed by HPLC. Establishing the TNF-αinduced malnutrition model of skeletal muscle cell, and observed cell morphologychanging. Furthermore,C2C12cells were divided into normal control group, TNF-αintervention group and medicated serum group. After TNF-α and medicated serumintervened6hours, detecting the expression of Ubiquitin、E214k、Atrogin-1、MuRF-1mRNA by real-time quantitative PCR(qRT-PCR). After TNF-α and medicated serumintervened72hours, detecting the expression of Ubiquitin、E214k、Atrogin-1、MuRF-1protein by western blot(WB).Results:There were gingsenoside Rb1,chlorogenic acid, ferulic acid, aconitine. Comparedto normal control group,after intervened by TNF-α, the myotubes fused delaying and themyotubes were significantly atrophy, and the muscular tube diameter significantly reduced.(P<0.05).The expression of Ubiquitin、E214k、Atrogin-1、MuRF-1mRNAand proteinwere significantly increased(P<0.05).Compared to TNF-α intervention group, afterintervened by medicated serum, the myotubes fused fast, myotubes were not obviously atrophy,and the muscular tube diameter were wider(P<0.05).The expression of Ubiquitin、E214k、 Atrogin-1、MuRF-1mRNA and proteinwere significantly reduced(P<0.05).Conclusion:Successfulling copyed malnutrition model of skeletal muscle cell by TNF-α. Baoyuan detoxofication decotion medicated serum could significantly reduced the expressionof Ubiquitin、 E214k、Atrogin-1、MuRF-1mRNA and protein. Illustrating Bao yuandetoxofication decotion probably inhibited skeletal muscle cell atrophy through UPPsignaling pathways, improving cachexia muscular atrophy status, providing new treatmentideas and effective prescription for treatmenting cancer cachexia.
