Objective: copy C26colon cancer cachexia mice model, the establishment ofevaluation method of cachexia model, the intervention of Bao Yuan detoxoficationdecoction on quality of life evaluation, research of cachexia model animal cell factorand the role of muscle atrophy, explore UPP pathways mediating mechanism ofcancer cachexia skeletal muscle atrophy.Methods: BALB/c mice right axillary subcutaneous inoculation C26colon cancercells, copying the cancer cachexia mice model. To calculate body quality drop rate,respiratory metabolism conversion rate (RER) were, heat yield H(1), H(2), to explorecachexia model evaluation method; Modeling successful40animals were randomlydivided into four groups, model control group (B), small Chinese medicineintervention measure group (C), Chinese medicine intervention in high dose group (D)and medroxyprogesterone acetate group (E), and normal control group (A), observethe general status in mice, record food intake, water quantity and quality, tumors,tumor weight, quality of the gastrocnemius muscle, toe long extensor quality, HEstaining observation of cell morphology changes of muscle fiber, and calculate thecell diameter of muscle fiber; ELISA method to detect the serum TNFalpha,IL-1,IL-6,IFN-gamma cytokine levels; Rt-pcr, immunohistochemical andWesternblot method determination of ubiquitin in muscle, E214K mRNA and proteinexpression.Results: copy cachexia model, the establishment of cancer cachexia modelevaluation method: general status, food intake, water quantity and quality decline rate,respiratory rate (RER) were, produce heat H(1), the index changes of H(2). After drug treatment, compared with group B, group C and group D food intake, water quantityand quality in mice were significantly increased (P<0.05), tumors and the tumorweight was significantly decreased (P<0.05), the quality of the gastrocnemius muscleand toe long extensor quality tends to increase (P>0.05), the cells of muscle fiberdiameter increased significantly (P<0.05), the TNF alpha,IL-1,IL-6, IFN-gammalevels of cytokines, ubiquitin, E214KmRNA and protein expression level decreasedobviously (P<0.05). Compared with group E, D group of tumors and the tumor weightsignificantly decreased (P<0.05), the quality of the gastrocnemius muscle and toe longextensor quality tends to increase (P>0.05), the cells of muscle fiber diameterincreased significantly (P<0.05), the TNF alpha, IL-1,IL-6,IFN-gamma levels ofcytokines, ubiquitin, E214KmRNA and protein expression level decreased obviously(P<0.05). Compared with group C, group D food intake in mice, water quantity,quality, quality, the toe of the gastrocnemius muscle cells of muscle fiber diameterand long extensor quality tends to increase (P>0.05),the tumors had product, tumorweight, TNF alpha,IL-1,IL-6,IFN-gamma levels of cytokines,ubiquitin, E214KmRNAand protein expression level decreased obviously (P<0.05).Conclusion:to establish the evaluation method of C26colon cancer cachexiamodel;Bao Yuan detoxofication decoction can improve the quality of cancer cachexiaanimal survival;reduce the content of TNF alpha,IL-1,IL-6,IFN-gammacytokine;reduce the content of Ubiquitin,E214K mRN TNA and protein expression.To objective the purpose of the thus suppressing skeletal muscle atrophy of cachexia.
