Objective: To establish the rats model of Liver-Qi depression and Spleen-Qi deficiency Syndrome with the gastroenteric motional function imbalance and its biological characteristics.Methods: This animal model is made by the method of confining the rats' movement and giving them vinegar by mouth. The animal model's syndrome is observed with quantitative and half-quantitative method. The animal model's intelligence level is measured according to water maze. The morphologic change of stomachs small intestine is watched according to HE staining. The ultrastructure change in the stomach and small intestine is observed with electron microscope. The gastroenteric motional function is measured according to the movement of semisolid substance. The lower thalamencephalon and jejunum level of 5-hydroxytryptamine (5-HT) and the plasma level of motilin(MTL) gastrin(Gas) neurotensin(NT) somatastin(SS) vasoactive intestinal polypeptide(VIP) and neuropeptide Y(NPY) are measured by radioimmunoassay(RIA). The electrophysiological change in the stomach and small intestine is noted by the multipurpose recorder.Result: (1)The rats model of Liver-Qi depression and Spleen-Qi deficiency Syndrome with the gastroenteric motional function imbalance have been established. Compared with the controlled rats, the general condition of the model rats is abnormal. Their symptoms are as followings: the puffed hair, the raspy voice with crouching together in the corner, the slow response with no struggle when seized, the decreasing appetite in 24 hours, the reducing movement, the falling body weight, flatulence with the loosen stool and dirty anus. The solid which is left in the stomach increases unusually (P < 0.05 ) . The small intestine's drive rate becomes slower than that of the controlled rats(P < 0.05 ).(2)The model rats have a series of biological characteristics.In comparison with the controlled group, the lower thalamencephalon and jejunum level of 5-HT increases (P < 0.05, P < 0.01) .The muscular tension of the stomach and small intestine decreases (P < 0.05). The gastroenteric hormone level is abnormal. The plasma level of VIP and NPY increases (P < 0.05,P < 0.01) and the SS increases also. The plasma has lower MTL level (P < 0.01). The plasma level of NT and Gas is lower also. The pathological and ultrastructure change in stomach and small intestine is obvious.Conclusions: Using the above mentioned method, we can establish the stable rat model of liver-Qi depression and Spleen-Qi deficiency Syndrome with the gastroenteric function imbalance. We also set up its biological characteristics. The lower thalamencephalon and jejunum levels of 5-HT increase. The muscular tension of the stomach and small intestine decreases. The gastroenteric hormone level is abnormal. The pathological and ultrastructure change in stomach, small intestine is obvious.
