Part 1 Clinical ivesttigation about the Ultrafine Compounding Particle of Kai Wei Jin Shi Tang on the regulation of gastroenteric motivationObjective: To initially observe the clinical therapeutic effect and safety about the ultrafine compounding particle of Kai Wei Jin Shi Tang on the patients with painful abdominal mass of weakness of the spleen and stomach type, then getting the message about its curative effect and effect on stomach intestine motivation of Chronic Superficial Gastritis (CSG),Functional Dyspepsia (FD) and Bile Reflux Esophagitis (BRE) .Method: Set up three groups that are times,1/2 times of the ultrafine compounding particle of Kai Wei Jin Shi Tang and the masculine comparison (Jian Wei Xiao Shi Pian), emplying the clinical research of trabant,single-blinds 1: 1: 1 comparison and multicentre. The patients with painful abdominal mass of weakness of the spleen and stomach type and one of those that CSG,FD,BRE are considered as the observation object. Estimate objectively the effect of symptoms and desease, then investigate the differences of indexs of gastric emptying time,time about barium reaching ileocecal junctions enteric emptying time and blood plasma motilin after two weeks of treatment. The last, initially observe the effect contrast about the ultrafine compounding particle of Kai Wei Jin Shi Tang on the patients with CSG,FD and BRE.Results: 7 of 120 patiens are shed and rejected, while 113 patients are actually finishied and statistically analyzed (37 for ultrafine particle times group,37 for ultrafine particle 1/2 times group,39 for comparison group) . It is no distinguished contrast in the information of those three groups before treatment and fine grouping equilibrate and comparability. The effect of symptoms in ultrafine particle times group,ultrafine particle 1/2 times group and comparison group is 86.49%,89.19% and 82.05%, while the desease effect of three groups is 78.38%,81.08% and 76.92%. There is no distinguished contrast in the effect among three groups (P>0.05) .It is obviously shorter in the indexs of gastric emptying time,time about barium reaching ileocecal junction and enteric emptying time after treatment among three groups (P<0.01), while comparing with others, the ultrafine particle 1/2 times group is shorter but no significant difference (P>0.05) . After treatment, it is obviously higher in the index of motilin (MTL) among three groups (P<0.05), while also no distinguished contrast (P>0.05) althout the ultrafine particle 1/2 times group is higher than others. There is distinguished contrast in the total effective,recovery and marked rate of ultrafine compounding particle of Kai Wei Jin Shi Tang on the patients with CSG comparing with on those with FD and BRE (P<0.05 or 0.01) . The incidence rate of adverse effect among those three groups is very low and has no significant difference, that is 8.11%,5.41% and 7.69% . There is no distinguished contrast in the indexs of blood routine,urine routine,stool routine,renal function,electrocardiogram, and so on among three groups before and after treatment.Conclusion: The ultrafine compounding particle of Kai Wei Jin Shi Tane has significantly therapeuticly effect on promoting stomach intestine motivation, and the effect of 1/2 times and times is identical. Meanwhile, its clinical safety is reliable. The well effect of ultrafine particle on such three common alimentary system deseases in order proper is from CSG,BRE to FD. Part 2 Effect of the Ultrafine Compounding Particle of Kai Wei Jin Shi Tang on the mice gastroenteric motivationObjective: To investigate the effect of the ultrafine compounding particle of Kai Wei Jin Shi Tang on gastric emptying,intestine facilitation of the normal mice and splenic asthenia mice.Method: Adopt the traditional broth of Kai Wei Jin Shi Tang and Jian Wei Xiao Shi Pian as comparison, chosing normal mice and Reserpine splenic asthenia mice as study target to observe the effect of ultrafine compounding particle of Kai Wei Jin Shi Tang on indexs of residual rate of stomach methyl orange,intestine weight and facilitation rate of intestine carbon particle, with different dose from times,1/2 times to 1/4 times.Results: (1)The result of normal mice: the residual rate of stomach methyl orange in different groups of ultrafine compounding particle of Kai Wei Jin Shi Tang is lower than normal group (P <0.05, P<0.01), the facilitation rate of intestine carbon particle in times and 1/2 times groups is higher than normal group (P <0.05, P<0.01), the intestine weight in 1/2 times group is weighter than normal one( P <0.05). While it is no distinguished contrast in such indexs between the traditional broth of Kai Wei Jin Shi Tang and normal group.(2)The result of splenic asthenia mice: The residual rate of stomach methyl orange of ultrafine compounding particle of Kai Wei Jin Shi Tang in both times and 1/2 times is higher than model group (P<0.01), while there is no significant difference between the 1/4 times and model one (P>0.05) . The intestine weight of traditional broth of Kai Wei Jin Shi Tang and 1/2 times,1/4 times of ultrafine compounding particle is weighter than model (P <0.05 or P<0.01), but that of times and model groups is no distinguished contrast (P>0.05) . The facilitation rate of intestine carbon particle of traditional broth of Kai Wei Jin Shi Tang and 1/2 times,1/4 times of ultrafine compounding particle groups is lower than model one (P <0.05 or P< 0.01), however, there is no significant difference between the times and model groups (P>0.05) .Conclusion: With regard to normal mice, the ultrafine compounding particle of Kai Wei Jin Shi Tang has significantly therapeudcly effect on promoting stomach intestine motivation, especially in 1/2 times. With regard to splenic asthenia mice, the ultrafine compounding particle of Kai Wei Jin Shi Tang in times dose has significantly therapeuticly effect on promoting gastric emptying, while has no effect on accommodating intestine motivation. The1/4 times group may slow intestine enterocinesia, but couldn't adjust stomach motivation. The 1/2 times and traditional broth of Kai Wei Jin Shi Tang entire amounts groups both has two—way indentical effect on promoting gastric emptying and slow intestine enterocinesia. Part 3 Mechanism research about the Ultrafine Compounding Particle of Kai Wei Jin Shi Tang on the regulation of gastroenteric forceObjective: To investigate the effect of the ultramicro compounding particle of Kai Wei Jin Shi Tang on conductive system of Ca2+- CaM -MLCK of gastroenteric smooth muscle cells of the splenic asthenia mice , to pursue it's possible mechanism of regulation of gastroenteric forse splenic asthenia mice.Method: Adopt the traditional broth of Kai Wei Jin Shi Tang and Jian Wei Xiao Shi Pian as comparison, by the methods of spectral photometer of fluorescence,PCR and immune molten to observe the effect of ultramicro compounding particle of Kai Wei Jin Shi Tang on activity of constriction or dilation of gastroenteric smooth muscle cells,the concen—tration of calcium ions within a cell and the expression of CaM Mrna and MLCK of the splenic asthenia mice.Results: (1)The affect of gastric sinus smooth muscle cells: the length of smooth muscle cells of gastric sinus in the model group is longer than normal group distinctly, the concentration of calcium ions within a cell is lower than normal group and the expression of CaM mRNA and MLCK in tissue decrease evidently (P<0.05) ; the length of cells in the group of Jian Wei Xiao Shi Pian,tradition and ultramicro compounding particle is shorter than model group clealy (P<0.01) , the concentration of calcium ions is higher than model group, the expression of CaM mRNA and MLCK is stronger than model group(P<0.05); the length of smooth muscle cells in the group of the ultramicro compounding particle is shorter than the group of Jian Wei Xiao Shi Pian obciously, the concentration of calcium ions and the expression of CaM mRNA and MLCK is higher than the group of Jian Wei Xiao Shi Pian ( P<0.05) ; while there is no significant difference between the group of ultramicro compounding particle and traditional one (P>0.05) ; (2)The affect of jejunum smooth muscle cells: the length of jejunum smooth muscle cells in the model group is shorter than normal group evidently, the concentration of calcium ions is higher than normal group (P<0.05 or < 0.01), the expression of CaM mRNA and MLCK in tissue increase evidently(P<0.05) ; the length of jejunum smooth muscle cells in the group of tradition and the ultramicro compounding particle is longer than model group obviously (P<0.01), the concentration of calcium ions is higher than model group,the expression of CaM mRNA and MLCK in tissue is lower than normal group (P<0.05 or <0.01) ; while there is no significant difference between the group of Jian Wei Xiao Shi Pian and model group (P> 0.05) ; the same to the group of tradition and the ultramicro compounding particle (P>0.05) .Conclusion: the ultramicro compounding particle of Kai Wei Jin Shi Tang can accelerates the contraction of gastroenteric smooth muscle cells and the dilation of jejunum smooth muscle cells, with relative mechanism possibly—the alteration of the concentration of calcium ions within a cell, the combination and vary of active of Ca2+-CaM,and further change of active of MLCK; all above effects of Kai Wei Jin Shi Tang is appropriate to the traditional broth but superior to Jian Wei Xiao Shi Pian.
