| Objective:Xiaoyao Powder was first recorded in " Prescriptions of the Bureau of Taiping People’s Welfare Pharmacy ",which was a classical prescription of traditional Chinese medicine for treating liver depression and spleen deficiency syndrome. The prescriptions of Xiaoyao Pill have two kinds,one includes the ginger and the other doesn’t.But the function of them is the same. Nobody knows the reasons for this phenomenon and nobody has found the relevant policy documents or reports about it.This paper discusses whether the ginger has some effect on liver depression and spleen deficiency syndrome and whether the prescriptions or the indications of Xiaoyao Pill need to be modified.Methods:①The objects were healthy adult male SD rats.〦xcept the negative control group, the other three groups were given the stress stimulation to establish the animal model randomly. The stimulation methods comprised the tail clamp, fasting, fasting,4℃ice-water bath and wet pad. The same stimulation can’t appear continuously. The rats in testing groups were given the test solution and the rats in model group and negative control group were given an equal volume water by intragastric administration.After21days,the blood was collected in abdominal aorta of rats and determined.③The evaluation index included signs, open-field test behavior (square crossing, standing, grooming)and’H-NMR metabonomics.Results:①In animal ethology, the number of square crossing and standing of rats in model group is different from that of rats in the negative control group, which indicated that the animal model had been established.The differences are significant between test groups and model group. It showed that the two different Xiaoyao Pills should have good intervention effect. There were no obvious differences between two test groups,which indicated that the overall effect of two prescription pills were not different.Meanwhile the weight in model rats grew slowly but the trends of weight growth of rats between two Xiaoyao groups and control group were the same.It supported the behavioral conclusion.②Analysis of PLS-DA showed that the overall callback functions of two prescription pills are same to metabolites of model rats.Compared with control group, the contents of8kinds of small molecular metabolites in the serum of model rats were different significantly(P<0.05). It indicated that these metabolites may be potential biomarkers of this syndrome. After drug administration, compared with the model group, the contents of8metabolites in the serum of rats in two Xiaoyao groups showed a callback to different degrees. All of them showed significant difference except for acetic acid. In addition,9kinds of small molecular metabolites between two test groups had significant difference.There were no confirmation of ownership and the metabolic pathways of these9metabolites now. The scientific research task wouldl be carried out in the future. It revealed that these metabolites may be potential biomarkers of intervention of ginger on liver depression and spleen deficiency syndrome.Conclusion:To sum up, this paper could draw some rational conclusions.Firstly, the results of behavioral experiments showed that Xiaoyao Pill had good therapeutic effect on liver depression and spleen deficiency syndrome and whether it contained the ginger had no significant influences on the overall effect. The results of metabonomics showed that the Xiaoyao Pill adjusted the contents of some small molecular metabolites on model rats and the ginger also influenced some small molecular metabolites on model rats. There were no enough evidences to prove that the prescriptions or the indications of Xiaoyao Pill must be modified. |
