Diarrhea type IBS overlapped FD syndrome belongs to "diarrhea", "pain", "full", "stomach", "acid reflux disease syndrome" category in TCM. The previous study has shown that liver depression and spleen deficiency is the most common type in patients with D-IBS overlapped FD. There has had diverse reports about establishing animal models of irritable bowel syndrome, FD and liver depression and spleen deficiency, but diarrhea type of IBS overlapped FD liver depression and spleen deficiency animal model has not been reported in China. Thus, We conducted the study of the establishment of D-IBS overlapped FD liver depression and spleen deficiency animal model and the research of its intervention mechanism in Chinese medicine with the support of national natural science foundation (Study on the mechanism of IBS overlapped FD liver spleen deficiency nerve-immune-endocrine disturbance, Item number:81173392).Experiment Ⅰ:The establishment of D-IBS overlapped FD liver depression and spleen deficiency animal modelTo confirm the rat model established by "clip tail stimulation+rhubarb lavage" was D-IBS overlapped FD liver depression and spleen deficiency through the status of the macroscopic view, behaviorchange, food ration, sugar consume, water consumption and weight change. Method:1, detect the macroscopic view, behavior change, food ration, sugar consume, water consumption and weight chang of the rat model; 2, analyze the data of open-field-test; 3, medicine test: explore the regulating effect on traditional Chinese medicine (TCM) s pain and definite pill and entrocoor dinatibiogen merged mosapride.Results:The reaction of model group changed from intense to bovine by clamped over time. Compared with normal control, significantly lower (P< 0.01) was detected of sucrose consumption in model group. The average locomotion velocity was statistically reduced (P<0.05) in open-field. Drug treatment has apparently adjustment to the indexes mentioned above.Conclusion:The method of tail clip+rhubarb lavage mode were used to simulate the pathological evolution process of the liver depression and spleendeficiency, and the syndrome of rat conformed to the symptoms in irritable bowel syndrome. Thus, we cloned the rat liver spleen deficiency model successfully.Experiment II:The changes of mental state, gastrointestinal hormone levels in D-IBS overlapped FD rat modelThe aim of the study was to investigate the psychological status, physical and biochemical indicators, and behavioral changes in the depression model of rat with diarrhea type irritable bowel syndrome overlapped with functional dyspepsia via open-field method as well as radioimmunoassay and immunohistochemical staining measurement. Methods:50 healthy male Wistar rats were randomly divided into five groups. Behavioral changes were observed in normal group and model group rats. Serum levels of vasoactine intrestinal peptide (VIP), somatostatin (SS),5-hydroxy tryptamine, endotoxin (ET) and interleukin-10 (IL-10), as well as interleukin-2 (IL-2) levels in the colon were examined.Results:The model rats had lower sugar consumption percentage (P<0.01), average activity speed in the open field (P<0.05), somatostatin levels (P<0.01), but higher vasoactive intestinal peptide levels (P<0.01) than the normal group. Compared with group D, endothelin levels of group B were significantly increased in the model rats (P<0.01).Conclusion:The severity of mental disorders was different between D-IBS and FD overlap syndrome models of rats, which was related to the increase in VIP and ET levels and the decrease in SS. There was close correlation between the lower gastrointestinal symptoms and modeling methods and disorders.Experiment III:Effects of Tongxieyaofang with Zhizhuwan on Diarrhea-predominant irritable bowel syndrome overlapped Functional DyspepsiaTo investigate the mechanism of Tongxieyaofang with Zhizhuwan in the treatment of rat model with diarrheogenic irritable bowel syndrome (D-IBS) overlapped functional dyspepsia (FD). Methods:To establish a comprehensive animal model of IBS overlapped FD, the normal control group, model group, western medicine, Chinese medicine low、middle and high dose groups were set up, observe behavior changes of rats, and the levels of plasma vasoactive intestinal peptide (VIP), omatostatin (SS), serotonin (5-HT) and endotoxin (ET) also had been detected.Results:The model rats that therapy of tongxieyaofang with zhizhuwan had higher sugar consumption percentage (P<0.01), average activity speed in the open field (P<0.05), somatostatin and serotonin levels (P<0.01), but lower vasoactive intestinal peptide levels (P <0.01) than the normal group. Tongxieyaofang with Zhizhuwan significantly improved the symptoms of diarrhea in rats.Conclusion:Tongxieyaofang with Zhizhuwan can antidepressant, inhibit the intestinal peristalsis significantly by elevating levels of plasma 5-HT, SS, reducing levels of plasma plasma VIP to play a role in treament of D-IBS overlapped FD.
