| Objective: By building an animal model of knee osteoarthritis in vivo,the effects of Danzikangxi Granules on TNF α,P38 MAPK and MMP13 of knee osteoarthritis were studied,and the mechanism of action of Danzikangxi granules in the treatment of knee osteoarthritis was discussed.Methods: Thirty-six healthy SD rats(♂)were randomly divided into 6 groups: blank control group,model group,Danzikangshi Granules low-dose group,Danzikangshi granules medium-dose group,Danzikangshi granules high-dose group and cecoxib capsules group,with 6rats in each group.The blank control group was not modeled,and the other groups were modeled by improved Hulth method for subsequent experiments.After successful modeling,each group received corresponding drug intervention.Danzikangshi Granules groups were given Danzikangshi granules,celecoxib group was given celecoxib dilution intragastric administration,blank control group and model group were given intragastric administration with distilled water,and the rats were sacrificed after continuous intervention for 8 weeks.The articular cartilage of rats was taken and the expression of P38 MAPK and TNF-α m RNA were detected by RT-PCR.The expression of TNF-α,P38 MAPK and MMP13 in peripheral blood was detected by ELISA.The pathological changes of cartilage tissue were observed by pathological staining.Results:Through the observation of experimental animals,it was found that the pathological behavior of rats in each dose group of Danzi Kangxi granule could be improved,which was less than that in the model group and showed a trend of improvement.By HE staining,safranin fast green staining and toluidine blue staining,it was found that the articular cartilage tissue of rats was significantly improved,and the cartilage surface,cells and cartilage structure were improved.By RT-PCR,it was found that the expression of p38 MAPK m RNA in the low dose group of Danzi Kangxi granule decreased,with statistical significance(P<0.01),TNF-α m RNA decreased,but no statistical significance(P>0.05);The expressions of p38 MAPK and TNF-α m RNA in the low,middle and high dose groups of Danzi Kangxi granule were statistically significant(P<0.01),and the medium dose had the best effect.The levels of p38 MAPK and MMP13 in the low,medium and high dose groups of Danzi Kangxi granule were statistically significant(P<0.05,P<0.01),Danzi Kangxi granule medium dose group had statistical significance on TNF-α content(P<0.01);The contents of p38 MAPK and MMP13 in the celecoxib group were statistically significant(P<0.05,P<0.01).Conclusion:Danzi Kangxi granule may inhibit the inflammatory response of knee osteoarthritis by regulating the p38 MAPK signaling pathway,so as to play a role in the treatment of knee osteoarthritis... |
