Objective:To observe the effect of danzikangxi Granule on the expression of p-IKK ? and the content of type II collagen in rats with knee osteoarthritis,and to explore the therapeutic effect of danzikangxi Granule on knee osteoarthritis.Methods: 50 male SD rats were randomly divided into 5 groups,10 rats in each group: blank group,sham operation group,control group A,control group B and experimental group.A and B control group and experimental group were treated with modified Hulth method to make KOA model.The sham operation group only simulated the operation process without damaging the ligament and cartilage structure of the knee joint.The blank group was not treated with modified Hulthl.The experimental group was given danzikangxi granule by gavage,the control group B was given diclofenac sodium enteric coated tablets by gavage;the blank group,sham operation group and control group A were given the same amount of normal saline by gavage.The levels of IL-1?,IL-18 and TNF-? in peripheral blood were detected by ELISA;the morphological changes of chondrocytes were observed by HE staining under light microscope;the expression of type II collagen was detected by SABC,The expression of p-ikk? protein was detected by Western blot,and the circumference of knee joint was measured before and after model establishment and drug intervention.Results: 1.There was no significant difference in the circumference of knee joint between the blank group and the sham operation group(P>0.05).The circumference of knee joint in the experimental group and the control group A and B was larger than that in the blank group and the sham operation group(P<0.05).The circumference of knee joint in the experimental group was smaller than that in the control group A and B,and the difference was statistically significant(P< 0.05).2.Under the light microscope,the articular cartilage sections of the four groups were observed.The morphology,cell number and matrix staining of the articular chondrocytes in the blank group and the sham operation group were basically normal.In control group A,the structure and arrangement of chondrocytes were disordered,the number of chondrocytes was significantly reduced,the matrix was dissolved,and some chondrocytes were necrotic.In control group B,the morphology of chondrocytes was complete,arranged orderly,the edge of cells was rough,some matrix was destroyed,and the staining was uneven.Compared with the control group A and B,the number of chondrocytes in the experimental group increased,and the matrix staining was uniform.3.The levels of IL-1 ?,IL-18,TNF-? and p-ikk ? in the blank group and sham operation group were lower than those in the experimental group and the control group,the difference was statistically significant(P<0.05);the levels of IL-1 ?,IL-18 and TNF-? in the experimental group were lower than those in the control group A and higher than those in the control group B,the difference was statistically significant(P<0.05);the expression of p-ikk? in the experimental group was significantly lower than that in the control group A and B.4.The expression of p-ikk? in the control group A The expression level of type II collagen in the experimental group was higher than that in the control group(P<0.05)Conclusion: Danzikangxi granule may reduce the expression of p-ikk ? protein,inhibit the activation of NF KB,reduce the content of IL-1?,IL-18 and TNF-?,slow down the inflammatory reaction,improve the synthesis and metabolism of type II collagen,delay the degeneration of articular cartilage and promote the synthesis and repair of cartilage,which has a positive effect on the treatment of knee osteoarthritis.
