Preeclampsia is an idiopathic disease during pregnancy,which is one of the important causes of increased maternal and preeclampsiarinatal mortality.Its clinical features are hypertension and proteinuria after 20 weeks of pregnancy.Common clinical treatments are sedative,antispasmodic,antihypertensive,anticoagulant,diuretic and other symptomatic treatments.The only way to cure is to terminate pregnancy,but this has a great adverse effect on the prognosis of perinatal infants.In recent years,there have been many reports on the treatment of preeclampsia using traditional Chinese medicine,especially combined Chinese and Western medicine.The combination of Chinese and western medicine for preeclampsia has the advantages of prolonging gestational weeks,reducing complications,and improving perinatal outcomes.Data mining technology can provide a basis for precise treatment of Chinese medicine by finding common laws and differences form from a large amount of medical data.It plays an important role in promoting medical academic research and accelerating the modernization of traditional medicine.Network pharmacology provides a new method for studying the mechanism of drug efficacy by systematically studying the complex network relationship between drugs and the body.The purpose of this thesis is to provide theoretical basis for the clinical treatment of the disease by studying the medication rules of traditional Chinese medicine for the treatment of preeclampsia through data mining,and studying the material basis and molecular mechanism of Chinese and Western medicine for preeclampsia through network pharmacology.Objective:1.Applying the data mining technical methods to explore the rules of Chinese medicine treatment of preeclampsia to provide certain ideas and methods for the fiuture treatment of this disease by Chinese medicine.2.Applying the network pharmacology technical methods to explore the material basis and the molecular mechanism of combined treatment of Chinese and Western medicine for preeclampsia according to the results of data mining and literature search.Methods:1.By collecting the relevant clinical literature of Chinese medicine treatment of preeclampsia in CNKI to establish a prescription medicine database,analyzed and summarized the frequency,efficacy,four natures,five flavors,channel tropism and association rules of traditional Chinese medicine.2.Based on the highest frequency Chinese herb obtained through data mining and the Western medicine that more reports in the literature in combination with this Chinese herb for the treatment of preeclampsia,network pharmacology was used to obtain the active ingredients and targets of the Chinese herb,the Western medicine targets and preeclampsia-related targets,and establish a compound-target network and interaction network between targets.Through GO enrichment analysis and KEGG pathway analysis studied the material basis and mechanism of Chinese and Western medicine for preeclampsia.Results:1.A total of 63 TCM prescriptions were included,among which 91 herbs were used and 14 herbs were used more than 12 times.The top three are salvia miltiorrhiza(SM),uncaria,poria.The major properties of TCM were cold.The major flavors of TCM were sweet.The major meridian tropism of TCM was liver meridian,heart meridian,spleen meridian and kidney meridian.In the association rule analysis,it was found that the drug pairs with the highest association strength were uncaria and gastrodia,followed by SM and Poria.2.Literature search founded that there were many clinical literature reports of SM combined with LMWH in the treatment of preeclampsia.Based on the network pharmacology technical methods,30 active components of SM were screened.Those compounds combined with LMWH acted on the 72 key targets such as STAT3,KNG1,TNF,VEGFA,IL6,FGFR1,EGFR,F10 and IL10.Furthermore,GO enrichment analysis and KEGG enrichment pathway analysis showed that SM combined with LMWH in preeclampsia involved positive regulation of cell proliferation,negative regulation of apoptosis process,positive regulation of MAP kinase activity,blood pressure regulation,positive regulation of nitric oxide biosynthesis process and other biological processes,and PI3K-Akt signaling pathway,TNF signaling pathway,HIF-1 signaling pathway,NF-κB signal pathway,VEGF signal pathway and other signal transduction pathways.Conclusion:1.Traditional Chinese medicine in the treatment of preeclampsia was based on the Pinggan Qianyang,and attached importance to promoting blood circulation and removing stasis,strengthening the spleen and kidney through data mining,which further revealed the law of medication and provided a reference for clinical treatment.2.Salvia miltiorrhiza combined with LMWH could control blood pressure and blood glucose,reduce inflammation,regulate immunity and so on by affecting blood vessel function,inflammatory reaction,glycolipid metabolism and immunity. |
