Objective:The division of Ephedra was studied in the early stage of our group,and the components of ephedra polysaccharide(bitter,cold),ephedra alkaloids(pungent,warm)and ephedra phenolic acid components(bitter,warm)were obtained.The classification of"property" and "taste" was studied by using system biology and pharmacological evaluation system of property and taste.In order to enrich the basic connotation of ephedra and its separated components’ property and taste theory and at the same time deepen the understanding of Ephedra’s water-promoting effect and its meridian.Based on the sanjiao edema model of different TCM syndromes for the first time,this paper discusses the water-promoting effect and the meridian of ephedra and its separated components.Method:(1)Establishment of the upper-jiao retained fluid rat mode:rats were injected with isopropyl adrenaline in skin,endotracheal intubation and cold stimulation.Test indicators:Serum protein and blood lipid(albumin,total protein,cholesterol,and triglyceride),weight and index of heart and lung organ,lung permeability index,lung dry-wet ratio,serum creatine kinase content,heart and lung HE staining.(2)Establishment of the spleen deficiency and dampness stagnancy rat model:rats were fed with high fat and low protein feed and weight-bearing swimming.Test indicators:Serum protein and lipid(albumin,total protein,cholesterol,triglyceride,high density lipoprotein cholesterol,and low density lipoprotein cholesterol),gastrointestinal function(gastric emptying rate,intestinal propulsion rate,gastrin,D-xylose excretion rate)and water load index.(3)Establishment of the nephrogenic edema rat model of kidney-yang deficiency syndrome:Hydrocortisone was injected intraperitoneally and Ariamycin was injected into tail vein.Test indicators:Serum protein and serum lipids(Albumin,total protein,total triglyceride,total cholesterol)renal function(serum creatinine,urea nitrogen),the target gland axis function index(T3,T4,E2,E2/T),the metabolism index(cAMP,cAMP/cGMP),urine protein and kidney HE staining.(4)Establishment of the nephrogenic edema rat model of kidney-yin deficiency syndrome:Rat gastric instillation of thyroxine tablets and injecting Ariamycin into the tail vein.Test indicators:Serum protein and serum lipids(Albumin,total protein,total triglyceride,total cholesterol)renal function(serum creatinine,urea nitrogen),the target gland axis function index(T3,T4,E2,E2/T),the metabolism index(cAMP,cAMP/cGMP),urine protein and kidney HE staining.Result:(1)Ephedra decoction had a strong water-promoting effect on Upper-jiao retained fluid model rats,which was consistent with the traditional Chinese medicine theory of ephedra acting on lung meridian.The alkaloid component of ephedra had obvious water-promoting effect on the Upper-jiao retained fluid model.(2)Ephedra decoction had a weak water-promoting effect on the model rats,which was consistent with the traditional Chinese medicine theory that ephedra did not return to the spleen meridian and liver meridian.The water-promoting effect of each component of Ephedra on the model rats was weak.Ephedra decoction and non-alkaloid components can improve serum lipid index in rats.(3)Ephedra decoction had a strong water-promoting effect on the model rats,which is consistent with the traditional Chinese medicine theory of ephedra acting on bladder meridian,The water-promoting effect of alkaloid component was very obvious,the effect of phenolic acid component was obvious,and the effect of polysaccharide component was weak.(4)Ephedra decoction had a weak water-promoting effect on the model rats.This maybe due to the symplectic warmth of ephedra,strong sweating power,consumption of body fluid,and Yin deficiency often results in loss of body fluid and blood.Ephedra polysaccharide has certain water-promoting effect on nephrogenic edema model of kidney-yin deficiency syndrome.Conclusion:(1)Ephedra alkaloid component is the pharmacodynamic material basis for the treatment of the Upper-jiao retained fluid model,it can be preliminarily inferred that the alkaloid component belongs to the lung meridian.(2)Ephedra decoction and non-alkaloid components can regulate the disorder of blood lipid,but the water-promoting effect on spleen asthenia edema is weak.(3)Ephedra alkaloid component and phenolic acid component are the pharmacodynamic material bases for treating nephrogenic edema model of kidney-yang deficiency syndrome,it can be preliminarily inferred that the alkaloid component and phenolic acid component belong to the bladder meridian.(4)Ephedra polysaccharide component is the pharmacodynamic material basis for the treatment of the nephrogenic edema rat model of kidney-yin deficiency syndrome.It can be preliminarily inferred that the polysaccharide component belongs to the bladder meridian.In this paper,the TCM syndromes edema model was used to evaluate the water-promoting effect and the meridian of the ephedra,and discuss the relationship between meridian and efficacy.In this study,we found out the separation components of ephedra with water-promoting effect on sanjiao edema models of different TCM syndromes,and explained the characteristics of water-promoting effect of ephedra and its components.The characteristics of the water-promoting effect basically reveal the meridian connotation of Ephedra and its components.This study is not only provides a theoretical basis for further elucidation of the substance basis of ephedra,but also provide an experimental basis and methodological discussion for the further study of components and monomers of traditional Chinese medicine. |
