Intestial tract is an important innate barrier for humans and animals to maintain the balance of the intestinal environment and to hinder pathogenic bacteria and toxins.Enteritis is one of the common diseases in dogs and cats in veterinary clinic due to the complex and numerous factors of intestinal infection.Usually,treatment for chronic enteritis is eliminating bacteria and removing inflammation.With the long-term use of antibiotics in the course of the disease,the body will inevitably show resistance,which will leads the weakened efficacy.At present,doctors usually add a variety of probiotics(such as Bifidobacterium animalis,Lactobacillus casei,etc.)to the gastrointestinal prescription diet for animals,in order to provide alternatives to traditional treatment.However,the current research shows that the effects of probiotics vary depending on the strain and dose,so that the benefits of prescription food containing probiotics cannot be exploited efficiently and quickly.Glutamine is a conditional and essential amino acid for the body.Under physiological or pathological conditions,it can prevent the translocation of bacteria and endotoxin,so as to ensure nitrogen and acid-base balance and glucose metabolism in the body.Glutamine is a necessary dietary supplements to maintain the integrity and barrier function of the gastrointestinal mucosa.Therefore,in order to investigate the therapeutic effects of glutamine in the body and intestines of dogs with chronic enteritis,glutamine is added to the gastrointestinal prescription food which has a large content of probiotics.In order to find a kind of diet which can be more efficiently in the aspect of reducing the inflammatory reaction in the body and intestinal tract,and solve the deficiency of ordinary prescription diet in adjuvant therapy.In this experiment,36 healthy Chineese dogs were randomly divided into 4 groups with9 dogs in each group,which were: glutamine experimental group(glutamine group),gastrointestinal prescription diet experimental group(prescription group),intestinal disease experimental group(enteropathy group)and healthy control group(control group).At 1-15 days after the start of the experiment,the experimental groups were fed full price adult dog diet(the ingredients only met the daily energy requirements)and aspirin(25 mg/Kg),while the control group was fed full price and adult dog diet.During the treatment period from 16 th to 26 th day(i.e.,from 1st to 11 th day after modeling establishment).The glutamine group was fed glutamine(200 mg/Kg)and Xinyuan gastrointestinal prescription food.Dogs in the prescription group were fed Xinyuan gastrointestinal prescription food.The healthy group and enteropathy group were fed full-price adult dog food.The feces of all dogs were collected on 2nd,6th and 10 th day after modeling.Fecal score was recorded and intestinal microflora was detected by 16 s RNA technique.Three dogs were randomly selected from each group on3 rd,7th and 11 th day after modeling,and performing duodenoscopy to check the duodenal injury.And then collecting serum to test level of antioxidant indicators(SOD,MDA,T-AOC).Take out duodenum,jejunum and ileum by operating end-to-end intestinal anastomosis,and test level of Ig M,Ig G,SIg A of intestinal tissues.The intestinal morphology was observed after H.E and PAS staining.At the same time,the expression of tight junction proteins(TJ protein)ZO-1 and Occlaudin in intestinal tract were detected through immunofluorescence technique in tissue sections.The result of experiment indicates that,in comparison with the enteropathy group and the prescription group,glutamine added to the prescription food could improve properties of excrement,reduce clinical symptoms such as diarrhea,and increase the species richness and species diversity in intestine.Enhancing the body’s antioxidant capacity by lowering the level of MDA and increasing the level of SOD and T-AOC.At the same time,strengthening immune response of intestinal mucosal by increasing the content of SIg A,Ig M,and Ig G.Glutamine can also improve intestinal morphological changes,such as villus growing and widening,V/C value increasing,mucosal muscule thickness decreasing,so as to improve intestinal digestion and absorption capacity.By reducing the quantity of IEL and GC in intestinal villi to improve the inflammation of intestinal tissue.Moreover,Glutamine can maintain the stability of intestinal mucosal barrier by increased expression of ZO-1 and Occlaudin.On the basis of canine enteritis with aspirin,above results suggest that combined feeding of glutamine and probiotics can ameliorate the pathological response and clinical symptoms caused by intestinal injury,and maintain the environmental homeostasis in the digestive tract,enhance the immunity and intestinal barrier function of body.Thus shorten the course of treatment.
