Porcine reproductive and respiratory syndrome(PRRS)is one of the most important diseases in the global pig industry.During the occurrence and development of PRRS,the clinical symptoms are very complex and the disease progresses rapidly.Currently,there is no established model for PRRS syndrome differentiation.In the process of large-scale breeding,individual syndrome differentiation is not suitable for clinical application,and the application of population syndrome differentiation and overall treatment can better meet the needs of modern disease treatment.It is necessary to practice and explore veterinary syndrome differentiation in PRRS.At the same time,many large-scale pig farms failed to achieve good control effects by using vaccines and antibiotics,and other methods.Inactivated and live attenuated vaccines available in production cannot provide adequate protection in most cases.Compared with vaccines and antibiotics,traditional Chinese medicine has unique advantages.Therefore,research and development of new veterinary drugs or traditional Chinese medicine feed additives to control PRRS are one of the ways to solve this problem.Therefore,this study carried out challenge experiment of piglets for syndrome differentiation and clinical evaluation of modified Yupingfeng powder combined with vaccine,and to comprehensively evaluate the regulatory effects of the combined use of veterinary drugs and vaccines on the body’s immunity,to provide the theoretical basis for the practice and popularization of traditional Chinese veterinary medicine.Specific research results are as follows:(1)Wei Qi Ying Xue syndrome differentiation and San Jiao syndrome differentiation theory based on traditional Chinese veterinary medicine,the observation of challenge piglets was carried out,and combined with the clinical manifestations and pathological changes of PRRS,the characteristics of TCM veterinary syndrome differentiation of PRRS were discussed.Through the challenge experiment of piglets,PRRSV challenge piglets were taken as the research object for syndrome differentiation analysis.PRRS syndromes were summarized into Wei syndrome,Qi syndrome,Ying syndrome,and Xue syndrome.The results showed that Wei-Qi syndrome appeared on 1~3d of the challenge,Ying syndrome appeared on 7 d,and Xue syndrome appeared on 14 d.(2)To explore the effects of modified Yupingfeng powder combined with vaccine on the average duration of wei-qi syndrome and the occurrence time of Yingxue syndrome in piglets,the process of PRRS transmission and clinical syndrome differentiation were summarized.The results were as follows,Average duration of Wei-Qi syndrome was as follows: combined group > vaccine group > challenge group;The occurrence time of the syndrome was as follows: challenge group >vaccine group > combination group.The results showed that the combination of modified Yupingfeng Powder and vaccine could greatly increase the duration of Wei syndrome and Qi syndrome,prevent the occurrence of transmutation and delay the occurrence of Ying syndrome and Xue syndrome.(3)In order to evaluate the regulatory effect of modified Yupingfeng powder combined with vaccine on the immunity of piglets,the serum contents of IFN-α,IFN-γ,IL-1β,TNF-α,IL-6,and IL-10 of piglets were detected by commercial ELISA kit of pigs,and the serum viral load of PRRSV was detected by fluorescence quantitative PCR at different times.The results showed that the modified Yupingfeng Powder and vaccine could promote the expression of natural immune cytokine IFN-α,reduce the level of immunosuppressor IL-10,reduce viral load,regulate the levels of TNF-α,IL-1β,and IL-6,which could not only enhance the innate immunity of the body but also prevent the overexpression of inflammatory factors and make it in a state of balance.
