| This study combined RT-PCRã€Western blot and immunohistology technique to investigate the dynamic changes of Steroidogenesis gene on the ovarian of sheep that collected each on 0ã€3ã€6ã€9ã€12d to approach the laws of Steroidogenesis gene changes through the estrous cycle in ovary of sheep, in order to pursue between energy metabolism and hormonesynthesis related genes of potential links. The results are as follows:It has been detected that the expression of StARã€3β-HSD showed a trend of rising and falling through 0-12d of esrous cycle and 9 d was highest(p< 0.05),But, steroidogenic factors 3β-HSD in ovary showed drastic changes across the estrous cycle ranging(p< 0.01).; But, CYP11 over the continuation of estrus cycle in 0ã€3ã€6ã€9ã€12d has a trend of rising in corpus luteum (CL) by the technology of RT-PCR. At the same time, the 3β-HSD and StAR protein was expression granulosa cells, oocytes, oocytes and corpus luteum at all levels in Follicular with corpus luteum and Follicular without corpus luteum, in particular were negative for CYP17A1 protein in oocytes. And, steroid hormone synthesis related genes StAR, P450 (CYP11A),3β-HSD mRNA relative expression level is consistent with the progesterone level in blood.The AMPK and Erkl/2 mitogen-activated protein kinase (MAPK) in CL showed drastic changes across the estrous cycle ranging a trend of rising and falling.Steroidogenesis related genes StAR, P450 (CYP 11 A),30-HSD take part in the process of sheep estrous cycle change, and the results positive correlation with AMPK and ERK1/2 pathway suggest that the steroid hormone synthesis is related to gene and follicular development and luteal function, and laid the foundation for further understanding the regulation mechanism of the sheep breeding process. |
