| Mongolian medicine “Sanhong Decoction” is composed of madder,loquat leaves and Purple grass with 10 g each,which has the effects of clearing lungs,relieving cough and fighting inflammation.This paper takes madder as the main research object,and analyzes the chemical components of the ethyl acetate parts of madder.At the same time,the volatile oil and trace elements of madder and loquat leaves from different origins are also analyzed and compared.This lays a certain foundation for the in-depth study of Mongolian Medicine “Sanhong Decoction”.The research contents are as follows:1.The madder was extracted with 75% ethanol under reflux,followed by extraction with petroleum ether and ethyl acetate to obtain the ethyl acetate fraction extract.After separation and purification by column chromatography and thin layer chromatography,6 compounds were obtained,which were:1,3,6-trihydroxy-2-methylanthraquinone,Dimethyl terephthalate,β –sitosterol,Dibutyl phthalate,1,3-dihydro-2hydroxymethylanthraquinone,1-hydroxy-2-methoxy-anthraqui none.2.The volatile oils of madder and loquat leaves from different origins were extracted by steam distillation,and the volatile oil components of madder and loquat leaves from different origins were analyzed and identified by gas chromatography-mass spectrometry(GC-MS).The experimental results show that the main compounds in the madder from Shaanxi,Hubei and Shandong are the same,the main compound is 5-(tert-butyl)-(1,1’-biphenyl)-2-ol,and the relative content is16.16%,17.31%,61.93%;the main compounds in loquat leaves from Zhejiang province,loquat leaves from Fujian province,and loquat leaves from Gansu province are the same,and the main compound is(E)-3,7,11-trimethyldodeca-1,6,10-trien-3-ol,the relative contents were 56.91%,58.17% and 57.24% respectively.3.The contents of trace elements potassium,sodium,calcium,magnesium,manganese,iron,copper,chromium,lead and nickel in madder and loquat leaves from different producing areas in Sanhong Decoction of Mongolian medicine were determined by air-acetylene flame atomic absorption spectrophotometry.The correlation coefficients of working curves of trace elements were0.9991-1.0000.According to the limit requirements of heavy metals and harmful elements in Hawthorn,the content of Pb is less than 2mg/kg.Cu content is less than10 mg/kg,according to the national food safety standards of Ni≤1.0 mg/kg;Cr 1.0mg/kg or less.The experimental results showed that the detection results of four heavy metal elements from different producing areas were all below the limit requirements.The above experiments provide the experimental basis and experimental evidence for the in-depth study of Mongolian“Sanhong Decoction”,which also lays the foundation for the subsequent identification of the origin of medicinal materials using fingerprint technology,and also provides a basis for the quality control of Mongolian “Sanhong Decoction”. |
