Electrospinning technology is a common method for preparing nanofibers,with simple equipment.Nanofibers made by elctrospinning has small diameter and high specific surface area.Hence,it is suitable for many fields."Haodele"powder is a traditional antibacterial Mongolian medicine composed of 14 herbs such as Phellodendron chinense,Sophora flavescens and Scutellaria baicalensis,etc.It has significant antibacterial effect in treating various skin diseases and is not easy to relapse and it has important potential for further development.However,due to the inconvenience and low utilization rate of the powder,it has certain shortcomings because it needs to be boiled and bathed.In this research,electrospinning technology was used to prepare drug-containing nanofibers with"Haodele"powder and aimed to improve its efficacy and convenience with the purpose of laying a foundation for the further development of patches.First,electrospinning was used to prepare drug-containing nanofibers,which were cross-linked with glutaraldehyde to improve water resistance.the surface morphology of drug-containing nanofibers was observed by scanning electron microscope,and the functional groups were identified by infrared spectroscopy.Then,rabbits were used to evaluate the skin irritation of drug-containing nanofibers.the efficacy of drug-containing nanofibers was analyzed through in vitro antibacterial experiments and compared with the antibacterial activity of the original powder.The drug-containing nanofibers were analyzed by high performance liquid chromatography(HPLC)to detect the content of berberine.Furthermore,the content of berberine,the active ingredient in the product,was tested and methodologically verified.Finally,liquid-mass spectrometry technology was used to detect the small molecule components in the"Haodele"powder and drug-containing nanofibers,and then the antibacterial mechanism was discussed.The results showed that the surface of the drug-containing fiber is uniform and smooth.With the increase of the drug content,the fiber diameter ranges from 165.6nm to 255.4nm,and the best dosing ratio is PVA:M=4:1(mass ratio).The infrared spectrum showed that the methyl peak appeared at 1632 cm-1 in the drug-containing fiber,which proved the drug was successfully loaded in the nanofiber.The new ether bond was generated at 1132 cm-1 to prove the successful cross-linking and improved water resistance.The results of the water absorption rate test found that the optimal cross-linking time is 6h,In this condition,the water absorption rate of drug-containing fiber reached 758%,and the solution loss rate was15%,which improved the water absorption effect of the drug-containing fiber.The skin irritation test results showed that the skin became red after a single administration,and there was no erythema,edema,itching and other reactions after multiple administrations.Further anlysis of tissue section analysis revealed that the skin was intact,indicating that the drug-containing fibers is safe to skin.Bacteriostatic experiments showed that the drug-containing nanofibers had obvious antibacterial effect on Staphylococcus aureus.When the drug content in the drug-containing fiber is only 6%of the powder group,the antibacterial ability can reached 73.1%of the powder group,indicating that the drug usage rate was effectively improved compared to powder.The HPLC method was used to establish a method for the determination of drug-containing fiber berberine.The precision,accuracy,durability,and stability were all in compliance with the pharmacopoeia specifications,laying a foundation for the establishment of quality standards.The average content of berberine in the drug-containing fiber was 1.71 mg/g,and the transfer rate of berberine from the powder to the fiber was 63.9%.After the detection of secondary metabolites of non-targeted drugs,1132small molecules were identified in the positive and negative ions mode.According to HMDB,Lipid Maps and KEGG database,it is predicted that there are 60 kinds of small molecules related to bacteriostasis.Among them,there are 33 kinds of small molecules are lower than that of the powder group and there are 18 kinds of small molecules are unchanged,while 9kinds of small molecules are up-regulated.The reason for the down-regulation may be that some small molecules are lost during the spinning process,The reason for the increase is that the powder is first stirred in the precursor solution for 12 hours to improve drug extraction rate.
