Effect Of MICT On The Expression Profile Of Gut Microbiome In Healthy Young Adults

Objective:In this study,through the intervention of 5-6 weeks of moderate intensity continuous training(MICT)in healthy young men,16S rRNA sequencing was performed on subjects’stool samples to determine the change trend of intestinal microflora structure and composition after 5-6 weeks of MICT intervention,as well as the impact of acute exercise training on intestinal microflora.Combined with the results of 16S rRNA sequencing,exercise cardiopulmonary function test,heart color ultrasound detection and body composition analysis,the influence of MICT on the body was discussed,and the beneficial mechanism of exercise was further explained from the perspective of flora homeostasis,so as to provide a theoretical basis for the application of MICT in public fitness.Methods:Eleven male subjects were randomly selected from Capital Institute of Physical Education who had no professional sports training experience within 2 years and were able to participate in this experiment.The exercise intensity was 60%VO2max,the exercise time was 30min,the exercise frequency was 3 times a week,and the exercise cycle was 5-6 weeks.Basic information,body composition,blood,cardiac ultrasound,exercise cardiopulmonary function and other checks were carried out twice within one week before training and one week after training respectively.The test conducted before training was called pre-test(QC),and the test conducted after training was called post-test(HC).Morning stool was collected within a week before training,48-72 hours after training and 24 hours after the first training.One person collected 3 stool samples at a time.Stool collected before training was called pre-test(QC),stool collected after training was called post-test(HC),stool collected after the first exercise training was called urgent test(JC).The collected feces were sequenced by 16S rRNA to explore the composition and function of the microbial flora in the subjects’gut.Result:(1)The results of body morphology test before and after 5-6 weeks MICT intervention showed that after exercise intervention,the body weight(p=0.262),BMI(p=0.279)and body fat percentage(p=0.030)of healthy adult males showed a decreasing trend.(2)16S rRNA sequencing results before and after 5-6 weeks MICT intervention showed that:After exercise intervention,the relative abundance of Firmicutes and Firmicutes/Bacteroidetes(F/B)in the intestinal flora of healthy adult males decreased,while the relative abundance of Bacteroidetes increased and Firmicutes/Bacteroidetes decreased.The relative abundance of Synergistetes(p=0.003)and Chloroflexi(p=0.018)was significantly different before and after exercise training.At family level,the relative abundance of Bacteroidaceae and Lactobacillaceae increased.At the genus level,the relative abundance of Bacteroides,Roseburia and Lactobacillus increased.There was no statistical significance in Alpha diversity and Beta diversity before and after exercise intervention.(3)16S rRNA sequencing before and after an acute MICT intervention showed that:After an acute exercise intervention,at the door level,the relative abundance of Bacteroidetes decreased,the relative abundance of Proteobacteria increased,and the ratio of F/B increased.The relative abundance of Synergistetes(p=0.012)had a significant difference before and after an acute exercise training.Division level,Synergistaceae(p=0.012),unclassified_Sorangiineae(p=0.048)and Desulfobulbaceae(p=0.036)significant difference on the relative abundance.Relative abundance of Butyricicoccus increases on the relative level.Desulfobulbus(p=0.014)and unclassified_Synergistaceae(p=0.048),unclassified_Sorangiineae(p=0.036)significant difference on the relative abundance.There was no statistical significance in Alpha diversity and Beta diversity before and after an acute MICT intervention.(4)CPET results before and after 5-6 weeks MICT intervention showed that:Before and after exercise intervention,Load max(p=0.017),VO2max(p=0.011),MET max(p=0.013),AT(p=0.007),VE max(p=0.0001)and MVV(p=0.0002)were significantly increased in healthy adult males.VO2/Kg max(p=0.398),VO2/HR max(p=0.249)and FCV1(p=0.252)had no significant difference.(5)The results of cardiac ultrasound examination before and after 5-6 weeks MICT intervention showed that GWI(p=0.010)and LVEF(p=0.039)were significantly increased before and after exercise intervention.GLS,GWE,GCW,GWW and SV were not significantly increased(p>0.05).(6)The correlation analysis between intestinal flora and body composition showed that:After exercise intervention,BMI was positively correlated with Blautia and F/B,and negatively correlated with Firmicutes,Bacteroidetes,Bacteroides,Alistipes and Roseburia,but there was no significant correlation(p>0.05).Body weight was positively correlated with Bacteroidetes,Bacteroides and Alistipes,and negatively correlated with Firmicutes,Blautia,Roseburia and F/B,but there was no significant correlation(p>0.05).(7)The correlation analysis results between intestinal flora and cardiopulmonary function showed that:After exercise intervention,VO2maxwas positively correlated with Firmicutes,Bacteroidetes,Prevotella,Lachnospiracea,Advenella,Prevotellaceae and Alcaligenaceae.It was negatively correlated with Bacteroides,but had no significant correlation(p>0.05).AT is negatively correlated with Firmicutes,Lachnospiracea,Prevotella,Advenella,Prevotellaceae and Alcaligenaceae,but positively correlated with Bacteroidete.Had a significant positive correlation with Bacteroides(r=0.83)(p=0.001).LVEF is positively correlated with Firmicutes,Alistipes,Roseburia,Ruminococcus,Butyricicoccus and Lactobacillus,while negatively correlated with Bacteroidetes(p>0.05).SV is positively correlated with Alistipes,Roseburia,Ruminococcus,Butyricicoccus and Lactobacillus,while negatively correlated with Bacteroides and Firmicutes(p>0.05).CLS is positively correlated with Bacteroidetes,Alistipes,Roseburia and Lactobacillus,and negatively correlated with Ruminococcus and Butyricicoccus,but has no significance(p>0.05).Was significantly negatively correlated with Firmicutes(r=-0.76)(p=0.03)LVEF is positively correlated with Firmicutes,Alistipes,Roseburia,Ruminococcus,Butyricicoccus and Lactobacillus,while negatively correlated with Bacteroidetes(p>0.05).SV is positively correlated with Alistipes,Roseburia,Ruminococcus,Butyricicoccus and Lactobacillus,while negatively correlated with Bacteroides and Firmicutes(p>0.05).CLS is positively correlated with Bacteroidetes,Alistipes,Roseburia and Lactobacillus,and negatively correlated with Ruminococcus and Butyricicoccus,but has no significance(p>0.05).Was significantly negatively correlated with Firmicutes(r=-0.76)(p=0.03).Conclusions:(1)The effects of 5-6 weeks of MICT and a single acute MICT on the community structure and diversity of human intestinal flora were different,but both of them changed the intestinal flora in the direction of health.The difference between the two is that 5-6weeks of MICT exercise training alters the composition of gut microbiota in healthy adult men and increases the relative abundance of probiotics,including Lactobacillus and Roseburia.However,an acute MICT exercise training changes the diversity of intestinal flora in healthy adult males and increases the relative abundance of Butyricicoccus.(2)5-6 weeks of MICT exercise training to improve the heart and lung function of healthy adult males,and play a role in weight loss,body shaping.(3)5-6 weeks of MICT exercise training changed the composition of subjects’intestinal flora,and the changes of intestinal flora were correlated with the enhancement of cardiopulmonary function and the improvement of body shape.
