Objective:to confirm the efficacy of Zhike Dingchuan plaster in the treatment of asthma model rats and to provide molecular biological basis for acupoint application in the treatment of bronchial asthma.Methods: 60 healthy male Wistar rats of SPF grade were randomly divided into 4 groups: normal group(n = 15),model group(n = 15),sham acupoint application group(n = 15)and acupoint application group(n = 15).Asthma models were established in all groups except the normal group.According to the experimental procedure,the rats in each group were given intervention treatment,and after the treatment,the rats were killed and samples were collected.1.The lung tissue of rats was stained with HE to observe and compare the infiltration of inflammatory cells in lung tissue,the exfoliation of bronchial epithelial cells,the degree of airway edema,hyperplasia and occlusion in each group,and to determine the improvement of physiological and pathological symptoms of asthma rats by Zhike Dingchuan paste.2.The expression of IgE in serum,IL-10 and IL-17 in lung tissue was detected by immunoadsorption(ELISA)method to judge the intervention effect of Zhike Dingchuan paste on asthma model rats.Results:1.The successful conclusion of the model is as follows:Compared with the normal rats,it was found that the asthma model rats had obvious changes in behavioral symptoms and pathological manifestations,and appeared abnormal symptoms such as asthma attack,which confirmed the success of the model.2.HE staining of lung tissue:In the normal group,the morphology of bronchial epithelial cells was intact,the morphology of alveoli was normal,and there was no edema,hyperplasia and inflammatory infiltration.The histomorphology of tissue cells changed in different degrees in model group,sham acupoint sticking group and acupoint sticking group,with different degrees of inflammatory cell invasion,bronchial stenosis and abnormal mucosal tissue.3.IgE expression assay were as follows:The expression of serum IgE in model group was significantly higher than that in normal group,the expression of IgE in sham acupoint group and acupoint sticking group was significantly lower than that in asthma model group,and the expression of IgE in acupoint sticking group was lower than that in sham acupoint sticking group.4.IL-10 expression assay were as follows:The expression of IL-10 in lung tissue of model group was lower than that of normal group,and the expression of IL-10 in sham acupoint group and acupoint sticking group was higher than that in asthma model group,and the expression of IL-10 in acupoint sticking group was higher than that in sham acupoint application group.5.IL-17 expression assay were as follows:The expression of IL-17 in lung tissue of model group was significantly higher than that of normal group,and the expression of IL-17 in lung tissue of rats in sham acupoint sticking group and acupoint sticking group decreased significantly compared with model group,and the expression of IL-17 in acupoint sticking group was significantly lower than that in sham acupoint sticking group(P <0.0l).Conclusion:1.Compare the behavior performance of rats in sham acupoint sticking group,acupoint sticking group and model group,and then compare the results of HE staining and conduit gene detection showed that the model of bronchial asthma rats was successful.2.The expression of IgE in serum of model rats increased significantly,the expression of IL-10 in lung tissue decreased significantly,and the expression of IL-17 increased significantly,which was correlated with behavioral and morphological symptoms and manifestations of model rats,suggesting that IgE,IL-10 and IL-17 were closely related to asthma attack.3.Zhike Dingchuan paste used in this experiment can relieve asthma symptoms,regulate immune balance and treat asthma by reducing the expression of IgE,increasing the expression of IL-10 in lung tissue and reducing the expression of IL-17 in lung tissue.
