Purpose: Based on the data mining technology to analyze the rule of prescription of compound prescription containing Astragalus in the treatment of Hashimoto's thyroiditis(HT),and explore the potential mechanism of Astragalus in the treatment of HT by network pharmacology.Material and method: The first part is the data mining research on the treatment of HT with Astragalus compound.We searched CNKI,Wanfang date,VIP,CBM and other Chinese databases for literatures published from the databases of establishment to October 2020.Screening the literatures that meet the criterias of inclusion and exclusion,and establish the database of HT treated with compound prescription containing Astragalus.SPSS statistics25.0 and SPSS modeler 18.0 were used for frequency analysis,association rule analysis and cluster analysis.The second part is the network pharmacology research of Astragalus in the treatment of HT.The TCMSP was used to screen the active components and related targets of Astragalus.The Genecards and OMIM databases were used to obtain the related targets of HT.The Cytoscape 3.7.2.was used to construct the "Astragalus-component-potential target"network diagram.The String11.0 database was used to construct the target protein interaction network.The potential targets of Astragalus in the treatment of HT were analyzed by KEGG signal pathway bioaccumulation through DAVID 6.8 databases.Result:1.(1)A total of 124 prescriptions were included in this study,and 172 traditional Chinese medicines were obtained except Astragalus.(2)Statistics of the frequency of compatible drugs:There were 22 high-frequency drugs(frequency?20)compatible with Astragalus.The top ten were Prunella,Poria cocos,Thunberg fritillary,Atractylodes macrocephala,Codonopsis pilosula,Licorice,Angelica sinensis,Longspur epimedium,Chinese thorowax root and Radix paeoniae alba.The most frequently used drugs were Qi tonifying drugs,followed by heat-clearing drugs,drugs resolving phlegm,drugs for promoting blood circulation and relieving blood stasis,drugs for diuretic and hygroscopic and drugs for regulating Qi. The properties of medicine were mostly warm,cold,and flat.The tastes of medicine were mostly sweet,bitter and acrid.The channel-tropisms of medicine were mostly liver,spleen and lung.(3)Association rules analysis: A total of 42 rules were generated.The most common connections are Astragalus + Poria cocos + Dried tangerine peel,Astragalus + Prunella + Radix scrophulariae,Astragalus + Thunberg fritillary + Radix scrophulariae,Astragalus + Thunberg fritillary + Oysters.(4)Cluster analysis: Four commonly used drug combinations with Astragalus were summarized.Class 1: Poria cocos,Atractylodes macrocephala,Codonopsis pilosula,Licorice,Rhizoma cyperi,Dried tangerine peel,Rhizome of chuanxiong,Pinellia ternata,Radix curcumae;Class 2: Prunella,Thunberg fritillary,Radix scrophulariae,Oysters,Radix Rehmanniae;Class 3: Angelica sinensis,Chinese thorowax root,Radix paeoniae alba,Curcuma zedoary;Class 4: Longspur epimedium,Prepared rehmannia glutinosa,Cornus officinalis,Chinese yam.2.Network pharmacology analysis: There were 17 active components of Astragalus that acting on 215 targets,and 1453 targets related to HT.The main active components of Astragalus in the treatment of HT may be Quercetin,Kaempferol,etc.The core targets may be TP53,JUN,AKT1,MAPK1,TNF,IL-6,etc.Enrichment analysis showed that Astragalus might play a role in HT through the following ways: throughing pathway in cancer,TNF signaling pathway,HIF-1 signaling pathway,Toll-like receptor signaling pathway,PI3 K Akt signaling pathway,T cell receptor signaling pathway and other multiple pathways.Conclusion:1.It is found that Astragalus in the treatment of HT is mostly combined with tonifying deficiency drugs,especially Qi tonifying drugs.And based on this emphasis is placed on the application of heat-clearing drugs,drugs resolving phlegm,drugs for promoting blood circulation and relieving blood stasis,drugs for diuretic and hygroscopic and drugs for regulating Qi to treat both the symptoms and the root causes.2.Multiple components of Astragalus play a role in the treatment of HT through multiple targets and pathways,which provides a reference for further explanation of its clinical mechanism.
