There are many kinds of metabolites in plasma,with large differences in nature and content.In this study,plasma samples from patients with coronary atherosclerotic heart and coronary atherosclerotic heart disease-type 2 diabetes mellitus were established,and the metabolomics high-resolution mass spectrometry data preprocessing process was established to screen the optimal solvent combination for plasma metabolite analysis.On this basis,the metabolic differences between healthy people and patients with coronary atherosclerotic heart and coronary atherosclerotic heart disease-type 2 diabetes mellitus were compared,and the plasma metabolic characteristics of coronary heart disease and coronary heart disease combined with type 2 diabetes were studied.The main research contents are as follows:1.Research on the pre-processing method of metabolomics high-resolution mass spectrometry dataAnalyze the characteristics of noise in metabolomics high-resolution mass spectrometry data,and establish metabolomics high-resolution mass spectrometry data pre-processing procedures,including MZmine 2.38 processing,matching the human metabolomics database,subtracting solvent blanks,and subtracting multiple peaks four Parts.The data pre-processing procedure established in this research is applied to the two sets of metabolite mixed standard data,data 1 deducts 97.40% noise,and data 2 deducts 96.56% noise.2.Study on the effect of different solvent combinations on the coverage of plasma metabolites15 different solvent combinations are selected to pre-process the plasma samples,and the data is filtered using the metabolomics high-resolution mass spectrometry data pre-processing process,based on the characteristics of the first and second mass spectrometry in the plasma,and the qualitative plasma metabolites To determine the optimal solvent combination for plasma metabolite extraction: protein precipitation reagent: methanol,reconstitution reagent: methanol.3.Study on the metabolic characteristics of patients with coronary heart disease and coronary heart disease with type 2 diabetesUsing ultra-high performance liquid chromatography-quadrupole-orbitrap mass spectrometry technology to carry out plasma metabolomics research in patients with coronary heart disease and coronary heart disease with type 2 diabetes to explore the metabolic characteristics of coronary atherosclerotic heart disease and coronary atherosclerotic heart disease-type 2 diabetes mellitus,In this study,78 endogenous metabolites in plasma were identified.On the basis of qualitative and quantitative metabolites,combined with methods such as partial least squares-discriminant analysis and variable importance projection,20 kinds of characteristic metabolites that distinguish healthy people and patients with coronary heart disease were screened,and 35 kinds of metabolites distinguished between coronary atherosclerotic heart disease and coronary atherosclerotic heart disease-type 2 diabetes mellitus patients,37 kinds of characteristic metabolites that distinguish healthy people and coronary atherosclerotic heart disease-type 2 diabetes mellitus.Metabolic pathway analysis results show that amino acid metabolism plays an important role in the physiological metabolism of these two types of patients,and fatty acid metabolism is likely to be related to coronary atherosclerotic heart disease-type 2 diabetes mellitus.
