In modern medicine,the treatment of hyperuricemia has attracted much attention.but there is no cure or drug.In recent years,traditional Chinese medicine has made some achievements in the treatment of this disease,but it still needs to be further developed.The research group carried out an experimental study on the intervention of acupuncture and moxibustion on hyperuricemia model rats,and concluded that acupuncture and moxibustion on Shu,Mu and Yuan acupoints in hyperuricemia model rats can reduce blood uric acid in different degrees,thus achieving the effect of preventing hyperuricemia.Based on the previous research results and considering the actual situation of clinical patients(fear of acupuncture,inability to receive treatment in hospital on a regular basis,etc.),the basic theory of traditional Chinese medicine was combined with the external treatment method of traditional Chinese medicine.In this study,acupoint application method was used to intervene the hyperuricemia model rats,and the intervention effect was observed in order to find considerable results.Objective:In this study through patches apply high uric acid hematic disease model rats of liver and spleen and kidney Shu and Mu acupoints on both sides(Gan Shu and Qi Men,Pi Shu and Zhang Men.Shen Shu and Jing Men),observe the intervention of high uric acid hematic disease model rats of SUA,SCr,BUN,OAT1 and ABCG2 expression level,the structure of the rat renal pathology and the influence of the general situation,and to evaluate,on this basis,discusses the intervention of patches in the apply method high uric acid hematic disease model of rat liver,spleen and kidney Shu raise hole the mechanism of action,for acupoint application method to treat high blood uric acid to provide more reference.It also provides scientific guidance for clinical acupoint application in the treatment of hyperuricemia in terms of drug formulation and acupoint selection.Methods:32 clean grade SD rats,male,were randomly divided into 4 groups:blank group,model group,Vaseline acupoint application group,drug acupoint application group,8 rats in each group.From the beginning of the experiment,rats in the three groups other than the blank group were given 10ml/kg potassium oxonate 2g/(kg·d)for modeling to prepare hyperuricemia model rats.In the blank group,the same volume of normal saline was administered once a day for 10 consecutive days.After 10 days,blood was taken from the tail vein of all the rats,and the SUA level of the rats was measured with automatic biochemical analyzer.Compared with the blank group,the SUA level of the other three groups was significantly increased(P<0.01),indicating that the HUA model was successfully prepared.Vaseline paste and drug paste were prepared by ourselves.After the intervention,the rats in the 4 groups were given once a day instead of once every other day.The blank group was fed normally without any intervention.The model group was fed normally with appropriate grasping and pressing without any other intervention.Vaseline acupoint application group and drug group were applied respectively with vaseline and drug stick on both sides of rat Gan Shu and Qi Men,Pi Shu and Zhang Men,Shen Shu and Jing Men cave and once per day,6 h every time,every time remove the post,with warm water to wipe,five times for a period of treatment,the rest 1 d after completion of each course,a total of three courses,18 days.After the intervention,SUA,SCr and BUN were measured by automatic biochemical analyzer.Immunohistochemical method was used to detect the positive expression levels of 0AT1 and ABCG2 in rat kidney and the pathological structure changes.Results:After statistical analysis of the collected data by SPSS20.0 software,the results were as follows:on the tenth day of modeling,compared with the blank group,the SUA level of the other three groups of rats was significantly P<0.01,and P<0.05 was found in the symptoms of darkening of hair color,listless mental state,and decreased body mass.Applied after the intervention,compared with the blank group,model group and vaseline meridian application group rats SUA and BUN level of P<0.01 or P<0.05,OAT1 and ABCG2 expression level of P<0.05 or P<0.01,then watch model group rats renal tubular atrophy,expansion,atrophy of renal tubular epithelial cells visible,fall off,irregular arrangement,urate crystals can be seen in the lumen,in interstitial inflammatory cell infiltration;With the model group and vaseline meridian application group,drug acupoint application group rats SUA,BUN level of P<0.01,OAT1 expression level was P<0,05 or P<0.01,the ABCG2 expression levels and its synchronization state,then observe the pathological damage degree and lighter,renal tubular expansion,epithelial cells atrophy improved,uric acid salt and inflammatory cell infiltration is not obvious,the color of rats,the mental state is on the mend.There was no significant difference in SCr levels in each group.Conclusion:The application of Shu and Mu acupoints on liver,spleen and kidney drug application intervention method can effectively reduce the SUA level of hyperuricemia rat model,the mechanism may be through increasing the protein expression of OAT1 and ABCG2 in rat kidney tissue,promote the excretion of uric acid through renal tubular,reduce blood uric acid,and reduce the damage to the kidney.
