With the changes of modern human diet structure,living habits and the sharp increease in life pressure.The incidence rate of hyperuricemia is increasing year by year,showing a younger trend.Hyperuricemia is easy to be ignored by patients because it has no obvious symptoms in the early stage,but it is closely related to gout,hypertension,coronary heart disease,chronic kidney disease and other diseases.The World Health Organization has included hyperuricemia in the category of metabolic syndrome.At present,the commonly used drugs for hyperuricemia have obvious side effects,cannot to be taken for a long time.By consulting a large number of documents,we found that acupuncture and traditional Chinese medicine have obvious therapeutic effect on hyperuricemia.Acupoint application is a traditional external treatment of traditional Chinese medicine,which integrates acupoint and traditional Chinese medicine treatment It has definite and mild effect,simple and safe.But there are few researches on acupoint application for hyperuricemiaObjective:In this experiment,the hyperuricemia rat model was made by intragastric administration of potassium oxyzinate.The observation was made by comparing the acupoint application of acid-lowing patch with that of false patch.The biochemical indexes of blood and urine,the excretion index of uric acid,the pathological changes of kidney issue,the expression of renal reabsorption transporter including URAT1 and GLUT9 were observed before and after treatment.To explore whether acupoint application can inhibit the high expression of URAT1 and GLUT9,reduce the reabsorption of uric acid in the kidney,promote the excretion of uric acid,reduce the accumulation of blood uric acid and renal uric acid.Providing new ideas and experimental basis for clinical treatment of hyperuricemia.Methods:At first,referring to the relevant literature on hyperuricemia and gout,combined with the guidance of the tutor,the composition of acid-lowing patch(ingredients:Shudi,Zexie,Chaihu,Shaoyao,Baizhu,Fuling,Baijiezi)and the acupoint of patch(Shu acupoint of liver,spleen,kidney,including both sides of Ganshu,Jimen,Pishu,zhangmen,Shenshu,Jingmen)were formulated.In this experiment,thirty-two adult male SD rats were randomly divided into four groups:blank group,model group,false Acupoint Application group and Acupoint Application group with 8 rats in each group.After one week adaptive feeding,the model group,the false Acupoint Application group and the Acupoint Application group were all given potassium oxyzinate to replicate hyperuricemia model,while the blank group was given normal saline at the same time.From the 11th day of modeling,the blank group was fed normally without any intervention,model group was take appropriate grasp,while false application group and acupoint application group were grasp,and no other intervention.the acupoint application group used acid-lowing patch to apply to the Shu-Mu point of liver,spleen and kidney for 5 days in a row,which was a course of treatment.The rest between the courses was 1 day,a total of 3 courses were intervened.The sham application group used the fake patch made of Vaseline,and its application method and time were the same as the acupoint application group.At the end of the treatment,24 hours urine was collected,5 ml blood was collected from abdominal aorta and left kidney was taken.The SUA and SCr were measured by full-automatic biochemical analyzer.According to the formula,the indexes of uric acid excretion included:24hUUA,FEUA,CUr.HE staining was used to observe the pathological changes of kidney and immunity The expression of URAT1 and GLUT9 were detected by histochemistry.Results:The results of this study are independent sample t-test,nonparametric test and one-way ANOVA test data.Before treatment,the SUA of model group,false application group and acupoint application group was significantly higher than that of blank group(P<0.01).After treatment,compared with the blank group,SUA in the model group increased significantly(P<0.01),24hUUA,FEUA,CUr decreased significantly(P<0.01),the expression of URAT1 and GLUT9 increased significantly(P<0.05).Under light microscope,appeared morphological changes of kidney and a large number of uric acid crystals.Compared with the model group,the expression of SUA,24hUUA,FEUA,CUr,URAT1 and GLUT9 in the false Acupoint Application group were not significantly different.Compared with the model group and the false Acupoint Application group,the SUA in the acupoint application group was significantly reduced,24hUUA,FEUA,CUr,was significantly increased(P<0.01),the expression of URAT1 and GLUT9 was significantly reduced(P<0.05).Under the light microscope,the pathological changes of the kidney were not obvious,and the uric acid crystal was significantly reduced.Compared with the blank group and the model group,the SCr of the false Acupoint Application group and the Acupoint Application group decreased slightly(P<0.05).Conclusion:(1)Hyperuricemia model can be made by given potassium oxyzinate,and the hyperuricemia state of rats can be maintained by continuous given potassium oxyzinate.(2)Acupoint Application can reduce the SUA level of hyperuricemia model rats and has therapeutic effect on hyperuricemia;(3)Eliminating the interference of restraint and other factors,Acupoint Application can inhibit the expression of reabsorption transporters URAT1 and GLUT9 in renal tubules of hyperuricemia model rats,inhibit the reabsorption process of uric acid in renal excretion,promote the excretion of uric acid through the kidney,reduce the accumulation of uric acid in the blood and kidney and reduce the damage to the kidney.
