Objective:GC-MS analysis technology is used to detect the abundance and content of metabolites in the serum.By comparing the metabolite characteristics of each group of animals,the characteristic metabolic markers of knee osteoarthritis are obtained,which provides new information for the diagnosis of knee osteoarthritis.Ideas.At the same time,explore the effect of iron-clad massage cream combined with scraping method(medicine scraping method)on knee osteoarthritis,and its effect on characteristic metabolites,and clarify its mechanism of action in the treatment of knee osteoarthritis.Methods:The random number table method was used to randomly grab a blank group of rabbits from 30 experimental rabbits,a total of 6 rabbits,and the remaining 24 rabbits for modeling operations.The papain solution was injected into the rabbit's knee joint every 3 days for 1 week,and a total of 3 operations were performed.Two weeks after the completion of the operation,a model rabbit was randomly selected,and the model was verified by comparing the range of motion,swelling of the rabbit's knee joint,and the morphology of cartilage and synovial membrane.After successful modeling,the remaining model rabbits were randomly divided into model group,medicine scraping group,iron-clad gold group,scraping group,and then standard intervention.The five groups of rabbits were all fed standardly.After applying the medicine in the scraping group,the scraping operation was performed.After the operation was completed,it was kept for 1 hour and then wiped off;in the iron-clad gold group,the corresponding therapeutic amount of iron-clad gold massage cream was taken and evenly applied to the right The knee joint of the hind limb was kept for 1 hour and then wiped off;in the scraping method group,the skin of the right hind limb of the rabbit was moistened with water,and the scraping operation was performed.All interventions were performed every other day.Observe and record the general situation uniformly.After standard intervention for 1 month,blood is collected from the heart to prepare serum for metabolomics analysis.Random forest method is used to find different metabolites between groups,and articular cartilage and synovium are taken for morphological observation And score.Results:1.According to the modified Lequesne MG score:thearticular cartilage of the model group is slightly degenerated,compared with the score of the blank group,P<0.05.The Tiebaojin massage ointment group was basically normal.Compared with the model group,P<0.05.2.The KOA pathological model was successfully created by injecting papain solution into rabbit knee joints.A variety of metabolic differences were screened in the serum,and the characteristic erythrose,lactic acid,glucose,arachidonic acid,oxalic acid,and inositol were analyzed.It was found that the metabolites in the serum of rabbit KOA model changed dynamically.Conclusion:Based on metabonomics,the altered erythrose,lactic acid,glucose,arachidonic acid,oxalic acid,and inositol found in serum can be used as potential and biomarkers for the diagnosis of KOA,as well as markers for the therapeutic effect of traditional Chinese medicine on KOA.The treatment of knee osteoarthritis by medicinal scraping may be achieved by acting on the three pathways of pyruvate metabolism,arginine and proline metabolism,and glycolysis/gluconeogenesis.This may be a combination of iron-clad gold massage cream and scraping.One of the mechanisms for the treatment of KOA.
