The Effect Of Scraping Method On The Proliferation Of Chondrocytes And The Expression Of TRPV6 And Sox9 In Rabbit KOA Model

Objective: To observe the proliferation of chondrocytes and the expression of TRPV6 and Sox9 in KOA treated with ti-bo-jin massage plaster scraping,and to investigate the effect of ti-bo-jin massage plaster scraping on the proliferation of KOA chondrocytes and the expression of TRPV6 and Sox9.To provide experimental basis for the study of the pathogenesis of KOA and the mechanism of the external use of traditional Chinese medicine.Methods: 40 New Zealand rabbits were divided into(A)blank group,(B)model group,(C)scraping group,(D)tiebojin massage paste group,(E)tiebojin massage paste + scraping group.Blank group will not be any intervention,the remaining four groups on the articular cavity injection of papain technique after the success of the preparation of rabbit model,adopt corresponding intervention,ear margin venous air embolism after 20 days to be put to death animals,cultivation using enzymatic digestion culture method in rabbit knee joint cartilage cells,using toluidine blue staining identified chondrocytes,Western Blot method to detect TRPV6,COL-?,COL-?,Sox9,COL-? expression quantity,CCK-8 method to detect the cartilage cell proliferation.Results:In TRPV6,COL-?,COL-? on three kinds of protein expression,blank group compared with model group,with significant difference statistically significant(P <0.01);The scraping group,tiebao-jin massage cream group and the model group showed statistically significant differences(P<0.05).The difference between tiebao-jin massage cream +scraping group and model group was statistically significant(P<0.01).On Sox9,COL-? protein expression,the blank group compared with model group,with significant difference statistically significant(P<0.01);The scraping group,tiebao-jin massage cream group and the model groupshowed statistically significant differences(P<0.05).The difference between tiebao-jin massage cream + scraping group and model group was statistically significant(P<0.01).Cck-8 method was used to detect cell proliferation.At the 0d time point,the model group,scraping group,tiebojin massage paste group and tiebojin massage paste + scraping group showed no statistically significant differences among the four groups(P>0.05),while compared with the blank group,the differences were statistically significant(P<0.05).At the time points of 3d,7d and 10 d,there were statistically significant differences between the scraping group and the tiebojin massage paste group and the model group(P<0.05),and statistically significant differences between the tiebojin massage paste +scraping group and the model group(P<0.01).Conclusion:Tiebojin massage cream combined with curettage can promote the proliferation of KOA chondrocytes,up-regulate the expression levels of TRPV6 and Sox9,and delay the degeneration of KOA articular cartilage.
