Objective: Objective to study the effect of Wenyang Shengji ointment on wound healing of diabetic skin ulcer in rats by observing wound tissue section,detecting relevant indexes and evaluating wound healing,and to discuss the mechanism of Wenyang Shengji ointment in intervening wound healing of diabetic skin ulcer,so as to provide certain experimental basis for clinical treatment of diabetic skin ulcer guided by external treatment theory of traditional Chinese medicine.Methods:1.Diabetic rat model was establishedAfter fasting for 12 hours,the diabetic model was induced by a single rapid intraperitoneal injection of streptozotocin(STZ).After 3 days of injection,the blood glucose was measured in the morning.If the measured blood glucose concentration was higher than 16.7 mmol / L and accompanied with "three polydipsia"(polydipsia,polydipsia and polyuria),the diabetic model was established successfully.After successful modeling,the blood glucose was measured at 8 a.m.the next day,and those whose blood glucose was unqualified were eliminated.2.Wound modeling and groupingA round skin defect about 2cm in diameter was made on the back of diabetic rats.Then the rats were randomly divided into experimental group,control group and model group.The experimental group was given Wenyang Shengji ointment,the control group was given b FGF,and the model group was given 0.9% normal saline.3.Take materials and fix themOn the 3rd,7th,14 th and 21 st day after administration,the same number of animals in each group were randomly selected and killed by neck removal method.The wound tissue was separated and fixed with 10% formaldehyde solution for standby.4.Experimental observationImage Pro Plus 6.0 image analysis system was used to calculate the wound healing rate of diabetic skin ulcer rats.HE staining was used to observe the tissue inflammatory infiltration,the number of new capillaries and fibroblast proliferation.Immunohistochemistry was used to detect advanced glycation end products(ages),receptor for advanced glycation end products(RAGE)and nuclear factor kappa B(NF-? b)The expression levels of TNF-?,IL-8,b FGF and EGF were detected.Results:1.Wound healing: the wound healing rate of the experimental group and the control group was significantly better than that of the model group on the 7th,14 th and21st day after modeling(P < 0.05),especially in the experimental group.2.HE staining observation: on 3D,7d,14 d and 21 d after modeling,there were more inflammatory cells infiltration and less fibroblasts and capillaries formation in the wound of model group at different observation time points;inflammatory cells gradually decreased and fibroblasts and capillaries proliferation gradually increased in the wound of experimental group and control group,and the experimental group was better.3.Immunohistochemical staining method test results:(1)statistical results showed that at different time points after administration,compared with the model group,the content of ages in the experimental group and the control group were decreased(P < 0.05),and the expression of ages in the experimental group was lower than that in the control group(P < 0.05).(2)At different time points,the expression of rage in the experimental group and the control group was lower than that in the model group(P < 0.05),and the expression of rage in the experimental group was lower than that in the control group(P < 0.05).(3)At different time points,the content of NF-? B in the experimental group and the control group was lower than that in the model group(P < 0.05),and the expression of NF-? B in the experimental group was lower than that in the control group(P < 0.05).(4)At different time points,the TNF-? content in the experimental group and the control group was lower than that in the model group(P <0.05),and the TNF-? expression in the experimental group was lower than that in the control group(P < 0.05).(5)There was no significant difference in the expression level of IL-8 among the three groups on the 3rd day after administration(P > 0.05);however,on the 7th,14 th and 21 st day after administration,the content of IL-8 in the experimental group and the control group was significantly lower than that in the model group(P < 0.05);and the expression of IL-8 in the experimental group was lower than that in the control group(P < 0.05).(6)On the 3rd day after administration,the expression of b FGF in the control group was higher than that in the experimental group and the model group(P < 0.05),and there was no significant difference in the expression of b FGF between the experimental group and the model group(P > 0.05);on the 7th,14 th and 21 st day after administration,the content of b FGF in the experimental group was lower than that in the control group(P < 0.05),but significantly higher than that in the model group(P < 0.05).(7)The content of EGF in the experimental group and the control group was higher than that in the model group on 3d and 7d after administration(P < 0.05),and there was no significant difference in the content of b FGF between the experimental group and the control group(P > 0.05);on14d and 21 d after administration,the content of EGF in the experimental group was lower than that in the control group(P < 0.05),but significantly higher than that in the model group(P < 0.05).Conclusion: Wenyang Shengji ointment has a good effect in promoting the wound healing of diabetic skin ulcer,significantly improving the wound healing rate and shortening the healing time.This may be due to the fact that it can reduce the expression and binding of ages and rage in the wound tissue of diabetic rats,reduce the activation of NF-? B signal transduction pathway,reduce the secretion of inflammatory factors and increase the synthesis of growth factors,so as to improve the inflammatory state of the wound,stimulate the growth of granulation tissue and accelerate the wound healing...
