In this study,the preparation of dizziness model was caused by mechanical rotation,and established the focal incomplete cerebral ischemia model through filament method,connecting with mechanical rotation and stimulation,to copy ischemia vertigo model.Evaluating Xifeng Dingxuan particles pharmacological effects against ischemic vertigo at the same time.Xifeng Dingxuan particle dose tested were 8.64,4.32,2.16g crude drug/kg,equivalent to the clinical human dosage 12,6,3 times,respectively,in which 4.32g crude drug/kg for the rat with respect to clinical human equivalent dose.1.Establishment of rat model of Incomplete focal cerebral ischemiaThe study was intended to establish a clear site of ischemia,the degree of ischemia controllable,and efficient and stable rat model of incomplete focal cerebral ischemia by MCAO modified suture method.According to body weight of rats to choose the corresponding specification of suture,and packet into different groups through controlling the depth of the suture into internal carotid artery.Respectively,13mm,14mm and 15mm groups.It was to judge the suitability and plasticity of models by detecting the rate of change in blood flow of vestibular nucleus before and after modeling.Experimental results showde that it was found that the blood flow was reduced to around 24.41%through the change of vestibular nucleus tissue blood befor and after modeling.Exactly,our experiment was to achieve the purpose,and realized the controllment of vestibular nucleus tissue blood flow,to determine the scope of ischemia,but does not cause lesions infarct size.Further in accordance with a standard that packet suture into ICA14mm.2.The influence of Xifeng Dingxuan particles on escape time of rats stimulated by rotatingIn this study,rats orally administrated four days,taking the method of preparation of dizziness caused by mechanical rotation,the result showed that Xifeng Dingxuan particles 1.08,0.54,0.27g/kg could significantly shorten the escape time(compared with model group P<0.01),and made the escape time shorten to 33.40,8.40,22.66%.Then Xifeng Dingxuan particles can significantly improve symptoms of vertigo.3.The influence of Xifeng Dingxuan particles on escape time of rats incomplete focal cerebral ischemia stimulated by rotatingThis study taken to reduce the diameter of suture and the distance of embolism,resulting in incomplete focal cerebral ischemia rat model according to the method of modified suture MCAO.After the rats regained consciousness,giving medicine at 1 time per day and lasting 4 days.After the fourth day of administration 1 hour,and through the complex mechanical rotation stimulating on the model rats to reproduce the incomplete focal cerebral ischemia vertigo rat model.Then determined the escape latency of rats.The results showed that:a significant increase in latency of rats in model group after rotating stimulated(compared with the sham group,P<0.01).The rats of Xifeng Dingxuan particles 1.08,0.54,0.27g/kg group can diminish the escape latency to 50.67%,43.13%,50.03%(compared with model group P<0.05).Xifeng Dingxuan particles improved ischemia vertigo symptoms significantly.4.The influence of Xifeng Dingxuan particles on brain tissue injury of incomplete focal cerebral ischemia vertigo ratsThis study taken to reduce the diameter of suture and the distance of embolism,resulting in incomplete focal cerebral ischemia rat model according to the method of modified suture MCAO.Giving medicine 7 days,and after the latest day of administration 1 hour,then taken the right brain(injured side)from the brain tissue raphe 1.1mm-2.5mm,fixed into 10%formalin.In accordance with the conventional procedures to make rat brain sagittal paraffin,HE staining,and observed in microscope.The Ischemic lesions of model rats located in thalamus,hypothalamus,basal ganglia,internal capsule parts,under the soft membrane,Intraventricular,and brain parenchyma appeared significant congestion and hemorrhage.At the same time,there existed the congestion and hemorrhage near the ischemic lesion site,losing a large number of nerve cells,and glial cell proliferating.These symptomes meet with the pathological features of focal ischemic brain injury described in loiterature.Compared with model group,Xifeng Dingxuan particles 1.08,0.54,0.27g/kg could significantly reduce the damage caused by ischemia,showed that there appeare congestion and hemorrhage slightly under the soft membrane,intraventricular,brain parenchyma.a small number of nerve cells also lost.5.The influence of Xifeng Dingxuan particles on the protective effect of brain tissue of incomplete focal cerebral ischemia vertigo ratsIn this study,rats administered orally for 7 days,It taken to reduce the diameter of suture and the distance of embolism,resulting in incomplete focal cerebral ischemia rat model according to the method of modified suture MCAO.The results showed that:Xifeng Dingxuan particles 1.08,0.54,0.27g/kg coudle be inhibit the content of Lac,MDA,NO in different degrees and increased the Activities of LDH,SOD and NOS,Among them,Xifeng Dingxuan particles 0.54,0.27g/kg had obvious effect(compared with model group P<0.01).6.The influence of Xifeng Dingxuan particles on the vestibular nuclei blood flow of incomplete focal cerebral ischemia vertigo ratsIn this study,rats orally administered for three days,It taken to reduce the diameter of suture and the distance of embolism,resulting in incomplete focal cerebral ischemia rat model according to the method of modified suture MCAO.The results showed that:each dose of Xifeng Dingxuan particles coudle be inhibit the vestibular nuclei blood flow of modle rat in vary degrees,which Xifeng Dingxuan particles 1.08,0.27g/kg performed obvious effect(compared with model group P<0.01).
