Objective:Using middle cerebral artery ischemia/reperfusion model of SD rats as objects of research. The article discussed the protetcive effect of four different concentrations of hypertonic saline on cerebral ischemia and reperfusion and done a comparative analysis to explore the optimal concentration of hypertonic saline and provide experimental evidence for the treatment of cerebral Ischemia-reperfusion injury, so give full play to the potential value of hypertonic saline in the application of neurology and provide reference values for clinical application.Method:1. The preparation of acute focal cerebral ischemia and reperfusion model; 2. 40 successful model were choosed and randomly divided into four groups of 10 each, respectively, at 10 minutes before ischemia and 12 hours after reperfusion, intravenous 0.9% NaCI 4 ml/kg (A group),7.5% NaCI 4 ml/kg (B group),10% NaCI 4 ml/kg (C group); 23.5% NaCI 4 ml/kg (D group); 3. All rats were graded scores of neurobehavioral function at 2 hours after surgery anesthesia,6 hours and 24 hours after reperfusion, next, two rats in each group were randomly selected and their brains were removed after fixed by using normal saline and 40 g/L Paraformaldehyde to perfuse.Their brains were made of 2 mm thick coronal slices at the edge of optic chiasm leading for histopathological observation by conventional production, Seiichi hematoxylin and eosin staining. In the infarct area, with a high power field as a unit area, counting the number of neutrophil at a continuous five fields. At last, get the mean value. While the rest eight rats in each group were made TTC staining determination of infarction volume and brain water content; 4.To collect all the datas of each group and do comparative analysis.Results:The neurobehavioral scores and infarct volume was measured no significant difference between groups (P> 0.05), D group's brain water content is less than the other group (P<0.05), the infiltration of neutrophils at infraction was also no significant difference between groups (P> 0.05).Discussion:The dehydration of 23.5% Nacl is better than the other three concentrations Nacl. About the infarct volume of Ischemia-reperfusion brain, infarct inflammation and the score of neurobehavioral function, four concentrations of Nacl were no significant difference.
