| Research objectives:This study simulated the lifestyle of people with circadian rhythm reversed.Mice were raised in an environment with circadian rhythm reversed.Through intervention of high-intensity interval training and medium-intensity continuous training,the effects of two different kinds of exercise on the metabolism and regulation of fat in mice under circadian rhythm reversed condition were explored.It provides an effective exercise program to improve fat metabolism for people under the condition of circadian rhythm reversal for a long time.Research Methods:In this study,32 healthy male SPF C57BL/6 mice aged 6 weeks were selected and divided into normal circadian rhythm quiet group(N group),circadian rhythm reversed quiet group(D group),circadian rhythm reversed moderate intensity continuous training group(E group)and circadian rhythm reversed high intensity interval training group(H group).Mice in normal and quiet circadian rhythm group maintained circadian rhythm light synchronously with the outside world,that is,12 hours of light.12 The mice were dark;Mice in the circadian rhythm reversal quiet group,the circadian rhythm reversal moderate intensity continuous training group and the circadian rhythm reversal high intensity interval training group received circadian rhythm reversal once a week,that is,the first week of 12 hours of light.12 hours of darkness,and then the second week,the reverse of the day and night,12 hours of darkness;12 hours of light exposure,and the overall exercise intervention duration was 12 weeks.Mice in group E(8 mice)ran on the platform for 60 minutes at a speed of 15m/min,once a day,5 times a week.Mice in group H(8 mice)exercised for 2min at a speed of 27m/min every day,and exercised continuously for 1min at a speed of 8m/min in the interval period,and repeated 12 cycles.Twenty-four hours after the last training,the experimental mice were fasted,weighed and measured for body length,and 1ml of blood was collected.The epididymal adipoid tissue was removed and wiped off with filter paper to show blood stains,and then the wet weight was weighed.The contents of serum TG,LDL-C,HDL-C and FFA were measured by automatic biochemical analyzer.Serum melatonin and leptin concentrations were determined by ELISA.The protein expression levels of fatty acid synthase,hormone sensitive enzyme and fatty acid transporter enzyme were determined by Western Blot.Results:1.Compared with group N,the body weight of group D mice increased,and the difference was significant(P<0.05);After exercise intervention,the body weight of groups H and E decreased compared with that of group D,and the difference was significant(P<0.05).2.Compared with N group,wet weight of epididymal adipose tissue and abdominal fat body ratio in D group were increased,and the differences were significant(P<0.05).Compared with N group,wet weight of epididymal adipose tissue and abdominal fat body ratio in groups E and H were decreased,and the differences were significant(P<0.05).Compared with group D,wet weight of epididymal adipose tissue and abdominal fat body ratio in groups E and H were decreased,and the differences were very significant(P<0.01).3.Compared with group N,the HDL-C concentration in group D was decreased with significant difference(P<0.05);Compared with group N,the concentrations of LDL-C and TG in group D were significantly increased(P<0.01),and the concentrations of FFA in group D were significantly increased(P<0.05).Compared with group D,the HDL-C concentration in group E was increased,and the difference was significant(P<0.05),while the LDL-C,TG and FFA were decreased,and the difference was significant(P<0.05).Compared with group D,TG and LDL-C in group H were significantly decreased(P<0.01).FFA decreased and the difference was significant(P<0.05).4.Compared with N,the serum leptin concentration in group D was decreased,and the difference was significant(P<0.05).Compared with group D,leptin concentration in group E was increased,and the difference was significant(P<0.05).Compared with group N,the serum melatonin concentration in group D was increased,and the difference was significant(P<0.05).Compared with group D,the serum melatonin concentration in group E was decreased,and the difference was significant(P<0.05).5.Compared with group N,the expression level of Fas protein in epididymal adipose tissue of mice in group D was increased,and the difference was significant(P<0.05).Compared with group D,the expression levels of histone in group E and H were decreased,and the difference was significant(P<0.05).Compared with group N,the expression level of HSL protein in group D was decreased,and the difference was significant(P<0.05).Compared with group D,the expression level of histone E was increased,and the difference was significant(P<0.05).Compared with group D,the expression level of histone H was increased,and the difference was very significant(P<0.01).Compared with the N group,the expression level of histone H was increased and the difference was significant(P<0.05).Compared with N group,the expression level of CD36 protein in D group and E group was increased significantly(P<0.05).Compared with group N,the protein expression level of group H was significantly higher(P<0.01).Conclusion:1.12 weeks of circadian rhythm reversal resulted in weight gain,abnormal lipid levels,and changes in enzymes regulating fat metabolism.In this study,circadian rhythm reversal mice developed fat metabolism disorders.2.Both types of exercise can effectively improve the body weight,blood lipid levels and lipid metabolism-related regulatory enzymes in circadian rhythm reversed mice.3.There was no significant difference between the two kinds of exercise in the intervention effect of circadian rhythm reversal mice. |
