ObjectiveBased on the method of data mining,this paper analyzes the rules of ofessor Wu Wei’s prescriptions for coronary heart disease,sorts out the common syndrome types of coronary heart disease treated by Pr ofessor Wu Wei in clinic by using the idea of drug testing syndrome,and sums up the core prescription of Professor Wu Wei for coronary he art disease and the addition and subtraction of commonly used drugs.Using the method of network pharmacology,this paper analyzes the cor e prescription,and discusses the mechanism of treating coronary hear t disease from the modern molecular level,so as to provide reference for the future basic experimental research.MethodsTo collect the prescription data of coronary heart disease patie nts in Professor Wu Wei’s clinic from December 2018 to December 2020,and collect and sort out the prescriptions that meet the inclusio n criteria.The v2.5 version of Chinese medicine auxiliary platform is used for data mining.According to the statistics of the usage fr equency of single traditional Chinese medicine in prescriptions,the TCM syndrome types were summarized.Based on the association algori thm,the most commonly used drug association combinations and prescr iptions were selected by taking the confidence as the screening cond ition,and the core prescriptions were selected by combining the con fidence and support.The components and corresponding potential targets of each Chinese medicine in the core prescription were obtained through disease batman-tcm website,and the coronary heart disease related targets were retrieved through gene cards website using "coronary heart ".Through the intersection of the potential targets of traditional Chinese medicine and the targets related to coronary heart disease,as the related targets of the core prescription for the treatment of coronary heart disease.The "protein-protein" interaction relationship between targets was obtained through string website,and the interaction relationship was analyzed by topology using Cytoscape,and the core targets were selected by degree.The KEGG analysis ofthe core targets was carried out by David,and the BP analysis of the core targets was carried out by cluego.ResultsAfter screening and exclusion,a total of 400 prescriptions were obtained.Based on the analysis of the frequency of use,the most f requently used 10 traditional Chinese medicines were chuanxiong,Dan shen,Chishao,Huangqin,Honghua,Maodongqing,Dalbergia odorifera,Huangqi,jujube and Zhuru.Under the condition of confidence greater than 0.95,there were 56 pairs of drug combinations,mainly focusin g on the efficacy of clearing away heat and promoting blood circulat ion.The results of syndrome type analysis showed that the most comm on type was heat toxin and blood stasis,followed by qi deficiency a nd blood stasis,heart pulse stasis,liver and kidney deficiency,qi stagnation and blood stasis,phlegm turbidity block,phlegm and blo od stasis,Yang Qi deficiency and phlegm stagnation.Through the ass ociation algorithm,the drug combination with the highest support is selected as the core prescription,which is composed of chuanxiong,Danshen,Chishao,Honghua,Huangqin,Maodongqing,huangqi and Jiang xiang.After screening the active components of eight traditional Chine se medicines,148 components and 1390 related targets were obtained,and 397 targets were obtained.A total of 78 core targets,mainly T NF,were screened by the data analysis function of Cytoscape-α、 I L-6,NOS3,IGF-1,TGF-1,etc.The BP analysis of 78 core targets by cluego showed that they were mainly enriched in seven biological pro cesses: regulation of nitric oxide biosynthesis,active regulation o f cell response to insulin stimulation,positive regulation of osteo clast differentiation,regulation of chemokine biosynthesis,regulat ion of chronic inflammation,regulation of fever,and regulation of macrophage differentiation.The results of KEGG analysis of core nod es by David mainly focused on 10 coronary heart disease related path ways: PI3 K Akt pathway,MAPK pathway,toll like receptor pathway,JA K STAT pathway,NF-κ B pathway-κ B pathway,VEGF pathway,m TOR pathway,adipocytokine pathway,PPAR pathway-γ Pathway,TGF-β Ac cess.Conclusion1.In data mining,the top ten traditional Chinese medicines are all products of clearing heat and activating blood circulation,and the core prescription is the prescription of clearing heat and activating blood circulation and replenishing qi,which indicates that director Wu Wei often uses the method of clearing heat and activating blood circulation in the clinical treatment of coronary heart disease.2.The results of syndrome type analysis show that heat toxin and blood stasis are the most common etiology and pathogenesis of coronary heart disease in Lingnan area,and the rest are Qi deficiency,qi stagnation,phlegm turbidity and other factors.3.the results of network pharmacology show that core prescriptions can treat coronary heart disease through regulating inflammatory reaction,protecting and proliferating vascular endothelial cells,proliferation and migration of smooth muscle cells,and macrophage foam.
