Purpose:This study is based on the modern literature in the network database,relying on the "Traditional Chinese Medicine Inheritance Computing Platform(V3.0)" to systematically explore the medication rules of the syndrome of Qi deficiency and blood stasis in coronary heart disease,deeply understand the connotation of drug compatibility,and explore potential drug combinations.Provide more data support for the clinical treatment of coronary heart disease with Qi deficiency and blood stasis syndrome.Using network pharmacology,predict the potential material basis and mechanism of action of the core group of drugs "Astragalus-Salvia-Ligusticum chuanxiong" in the treatment of coronary heart disease from the molecular level,paving the way for follow-up experimental research.Material and method:Search the clinical research literature of TCM treatment of coronary heart disease Qi deficiency and blood syndrome from the establishment of the CNKI database to 2020.12.31,comply with the literature acceptance criteria,and input the traditional Chinese medicine prescriptions in the literature for the treatment of coronary heart disease Qi deficiency and blood stasis syndrome into the "Traditional Chinese Medicine Inheritance Computing Platform(V3.0)" for data mining and analysis,using frequency statistics,association rule analysis and cluster analysis(kmeans algorithm + regression simulation)to mine high-frequency drugs,core drugs,high-frequency drug combinations,core drug combinations and new prescriptions combination.Using the method of network pharmacology,screening the components and targets of the core group medicine "Astragalus-Salviae-Ligusticum Chuanxiong" excavated in the previous stage through the TCMSP and Drugbank databases,and screening the targets of coronary heart disease through the Gene Cards,OMIM and TTD databases,After standardizing the protein genes of drugs and disease targets by Uniprot,using VENNY for mapping,using STRING database for PPI network analysis,and using Cytoscape software to modify,perform GO enrichment analysis and KEGG enrichment analysis,and finally use Omic Share cloud The platform performs data visualization processing.Results:1.A total of 439 prescriptions of Chinese medicine were included in this study,and a total of213 medicines appeared.Using frequency statistics,it is concluded that in the clinical treatment of coronary heart disease with Qi deficiency and blood stasis syndrome,tonic drugs are the most commonly used,followed by activating blood and removing blood stasis drugs,supplemented by heat-clearing drugs,hemostatic drugs,anti-epidermal drugs,qi-regulating drugs,phlegm-relieving cough and asthma drugs Medicines for calming the liver and extinguishing wind,diuresis and dampness medicine,tranquilizers,etc.The four qi are mainly warm,followed by cold and flat.The five flavors are mostly sweet,bitter and pungent.Mainly belong to the liver meridian and heart meridian,followed by the spleen meridian and lung meridian.Through association rule analysis,12 pairs of high-frequency drug combinations were mined,6 core drugs: Astragalus,Danshen,Chuanxiong,Angelica,Red Peony,Safflower,and 20 pairs of core drug combinations.Through cluster analysis(kmeans algorithm + regression simulation),a total of 22 new recipes were mined.2.Based on network pharmacology,a total of 79 active ingredients in the core group of drugs "Astragalus-Danshen-Chuanxiong" for the treatment of coronary heart disease with Qi deficiency and blood stasis syndrome are predicted,including quercetin,kaempferol,luteolin,tanshinone IIA,7-O-methylisomucronulatol and other key active ingredients,there are 180 potential targets corresponding to drugs and diseases,mainly PTGS1,PTGS2,ADRB2,OPRM1,NCOA1 and other targets.The result of GO biological function enrichment analysis is the reaction to metal ions.Oxidative stress response,cell response to drugs,response to lipopolysaccharide,response to bacteria-derived molecules,response to steroid hormones,response to oxygen levels,response to external stimuli,etc.The results of KEGG pathway enrichment analysis are PI3K-Akt signaling pathway,TNF signaling pathway,IL-17 signaling pathway,Th17 cell differentiation,HIF-1 signaling pathway,etc.Conclusion:1.The basic principle of clinical treatment of coronary heart disease with Qi deficiency and blood stasis syndrome is to replenish qi as the core,promote blood circulation to remove blood stasis,and nourish blood,promote qi and relieve pain.Hemostatic drugs,anti-epidemic drugs,qi-regulating drugs,phlegm-relieving cough and asthma-relieving drugs,liver and wind-relieving drugs,diuresis and dampness drugs,tranquilizers,etc.2.The mechanism of action of the core group of drugs "Astragalus-Salvia-Chuanxiong" in intervention of coronary heart disease may involve the reaction to metal ions,the reaction to oxidative stress,the reaction of cells to drugs,the reaction to lipopolysaccharide,and the reaction to bacteria-derived molecules.Response,response to steroid hormones,response to oxygen levels,response to exogenous stimulation and other pathological processes,and PI3K-Akt signaling pathway,TNF signaling pathway,IL-17 signaling pathway,Th17 cell differentiation,HIF-1 signaling pathway,etc.Significant correlation.
