When using human surface contact devices,they will directly contact skin,mucous membrane or damaged surface.Such devices will be contaminated by pathogenic microorganisms,which will cause the user to infect various diseases and cause harm to the body.Contact lens is a contact instrument with mucous membrane.Due to its advantages of portability,vision correction and suitable for wearing during sports,contact lens has gradually replaced frame glasses and become a daily item in people’s life.In order to make the wearing of contact lens safe and effective,the wearer needs to regularly clean,disinfect,soak and other nursing steps of the lens,to keep the lens clean,and to prevent eyes from being infected.At present,the products which are used for disinfection of contact lens in the market generally have the problem of high content of disinfection components,and long-term use will produce certain biological risks,so it is very necessary to carry on the disinfection evaluation to it.In this paper,the disinfection effect of contact lens was evaluated in terms of bactericidal property and biocompatibility.Choose different components,different concentrations and different effective disinfection disinfection effect time in YY 0719.3-2009 "Ophthalmic optics.Contact lens care products.Part 3:Microbiological requirements and test methods for products and regiments for hygienic managements of contact" in direct germicidal test method to investigate different components at different concentrations of sterilization disinfection disinfection of the shortest time when required.Due to its good neutralizing effect,the D/E neutralizing broth of BD Company was selected as the neutralizing agent in this study.First,direct sterilization test was carried out.When the effective disinfectant was polyhexagenetformin(PHMB):the sample concentration of 1.0 PPM could meet the minimum sterilization requirements at 1.5h;At the concentration of 1.5 PPM,1.8 PPM and 2.0 PPM,the action time of 1h can reach the minimum sterilization requirements.The sample with a concentration of 5.0 PPM can meet the minimum sterilization requirement in 2.5min.When hydrogen peroxide was the effective disinfectant,the samples with concentration of 0.1%,0.2%and 0.3%all reached the minimum sterilization requirements in the action time of 30min.The results showed that the sample with PHMB concentration of 5.0 PPM had the best disinfection effect.Secondly,biological evaluation was carried out on all samples.When PHMB was the effective disinfectant,all samples were evaluated as non-cytotoxic according to the cytotoxic agar diffusion method.According to the MTT cytotoxicity test,the sample with a concentration of 1.0 PPM was evaluated as potentially cytotoxic at 30%,40%and 50%dilution.The concentration of 1.5 PPM was evaluated as potentially cytotoxic at 40%and 50%dilution.The samples at 1.8,2.0,and 5.0 PPM were potentially cytotoxic at all dilution concentrations.The results showed that the higher the concentration of PHMB,the greater the risk of potential cytotoxicity,and the better biocompatibility was evaluated when the concentration of PHMB was 1.0-1.5ppm.Hydrogen peroxide is highly oxidizing,and all the samples have a high risk of cytotoxicity.This type of product cannot be directly used after the contact lens is disinfected.Generally,certain operating procedures are required to completely neutralize hydrogen peroxide.To ensure that the above disinfection effect and biocompatibility test results are reliable,sterility tests were conducted on the samples involved,and the results showed that all samples met the requirements of sterility.In addition,this paper explores the application of flow cytometry in sterility detection.Compared with traditional methods,flow cytometry reduces the detection time of bacterial species by 40%,and has higher sensitivity,which can quickly and accurately detect the bacterial species within the range of 100cfu/mL,which can be used as an auxiliary of traditional methods to improve detection efficiency.At the same time,the flow cytometer can distinguish the microorganisms and other substances in the sample according to the different fluorescence color and the strength of the output signal,so as to further analyze and study the sample,which has certain scientific significance.In conclusion,the use of PHMB with a concentration of 1.5 PPM to disinfect the contact lens has a good evaluation of disinfection effect and biocompatibility.In addition,the shorter disinfection time also has a certain convenience.Human surface contact devices need to be disinfected before repeated use in order to meet the standard of use.Biological risks of these disinfectants to users should be fully considered when evaluating the disinfection effect.Therefore,this paper takes the disinfection evaluation of contact lens as the starting point to carry out the research,aiming to provide reference for the disinfection effect evaluation of medical devices contacting human surface,and further establish a disinfection scheme with standard sterilization effect and good biosafety,so as to provide data support for the research and development of related products.
