| 1.ObjectiveTo observe the effects of Potentilla Discolor Bunge on blood glucose,blood lipids and insulin resistance in type 2 diabetic patients and rats.2.MethodsSixty patients who met the inclusion criteria were randomly divided into control group and study group.The control group was given basic hypoglycemic drugs,study group was additionally treated with Potentilla Discolor Bunge Granules based on the drug used in the control group,and both groups were observed for 8 weeks.The FBG,2h PG,FINS,TG,TC and Hb A1c were detected and the resistance index(HOMR-IR)and Insulin Sensitivity Index(ISI)was calculated on patients of both groups before and after the treatment.Forty male SD rats were fed adaptively for one week,and then randomly divided into normal control group(8 rats)and model group(32 rats).The model rats were fed with high-fat and high-sugar for 4 weeks,combined with streptozotocin 30 mg·kg-1Intraperitoneal injection was used to replicate the type 2 diabetes rat model.According to the different treatment of Flavonoids from Potentilla Discolor Bunge of 160 mg(kg·d)-1,Pioglitazone of 1.95 mg(kg·d)-1and same amount of normal saline,the rats were randomly divided into three groups of Flavonoids from Potentilla Discolor Bunge group,Pioglitazone group and model control group.The normal control group was fed with ordinary feed for 4 weeks,and was injected intraperitoneally with an equal volume of citrate buffer solution,and the same volume of normal saline was intragastrically administered.After 4 weeks feed,abdominal aortic blood were collected,and later the Fasting Blood Glucose(FBG),Fasting Insulin(FINS),Triglyceride(TG),Total Cholesterol(TC)of each rat was detected and the ISI was calculated.3.Results(1)The clinical study found that the decreasing trend of BMI,FBG,Hb A1c,TG,TC in the study group was more obvious than that in the control group,but there was no statistical significance(P>0.05).The amelioration trend of study group was obviously better than the control group 2h PG、FINS、HOMA-IR and ISI(P>0.05).(2)Compared with the normal control group,the weight and FBG of the SD rat model control group were increased significantly(P<0.05).After 4 weeks of drug intervention,compared with a model control group,glucose and lipid metabolism disorder and insulin sensitivity were significantly improved in Flavonoids from Potentilla Discolor Bunge group(P<0.05).4.Conclusion(1)Potentilla Discolor Bunge can effectively reduce the level of 2h PG,FINS and HOMA-IR,and improve the level of ISI in patients with Type 2 Diabetes Mellitus.(2)Flavonoids from the Potentilla Discolor Bunge can significantly improve glucose and lipid metabolism disorder and enhance insulin sensitivity in type 2 diabetic rats. |
