We choosed Potentilla Disclor Bunge that was used by many DM people. The aim was to study the hypoglycemic effect of Potentilla Disclor Bunge, searched after hypoglycemic ingredients in Potentilla Disclor Bunge,and optimized the extraction and purification techniques of the active units of the Potentilla Disclor Bunge and also studied its pharmacodynamics by different experiments.We first from the raw material for medicine shape;the micro distinction, the physics and chemistry distinction and so on several aspects have done the pharmacognosy research.Has made the system pretesting to the chemical composition.Established the dentification method of Potentilla Disclor Bunge.Use spectrophotometric method to detect the total counts of flavonoids components was established.It provides foundation for the extraction and purification techniques of the active components in Potentilla Disclor Bunge.The extracts of Potentilla discolor Bunge were processed by dissolvent subsection extracting.Then adopt hyperglycemic model by mice to study the pharmacological effect of the different extracts from Potentilla discolor Bunge.The total flavonoids in different extracts from Potentilla discolor Bge. was determined by UV-spectrophotometry.Determined that flavonoids is the active component of reducing glycemia function in Potentilla discolor Bunge.The total count of the active component was detected and the orthogonal design was applied to inspect the elution concentration of the ethanol,menstruum volume,extraction duration,extraction frequency so as to determine the best purification techniques of the Potentilla Disclor Bunge.The best purification techniques were as follow:70%ethanol six times volume recirculated two times,each time continued two hours.The macroporous adsorptive resins were taken to purify the active units of the Potentilla Disclor Bunge,we investigated the the resin types,the example amounts,the water elution solvent and volume,the elution flow rate to determine the best purification techniques:D101 type macroporous adsorptive resins,ethanol and the extracted things(ml/g) proportion was 20:1,3BV water to elute,then 2BV70%ethanol to elute,elution flow rate was 1.25 ml/cm~2.min.The high lipoids foods were fed then the streptozotocin in small dose was given to make diabetes models in rats.After that,the active components in the Potentilla discolor Bunge were administrated intragastrically.Then the blood glucose,the insulin level in serum,the glucose tolerance,activity of SOD and the amount of MDA were detected to observe its antidiabetic effects.We have proved that the active components in the Potentilla discolor Bunge could decrease the blood glucose and increase the insulin level in serum,improve the glucose tolerance,clearancing free radicals and process of anti-lipid peroxidation,promoting the rehabilitation of islet.
