| Background:Diabetes mellitus(DM)is a common endocrine and metabolic disease,which is characterized by absolute or relative deficiency of insulin,which causes disorder of glucose,fat and protein metabolism.It is characterized by hyperglycemia.Long term hyperglycemia will lead to chronic damage and dysfunction of various tissues and organs,especially eyes,kidneys,heart,blood vessels and nerves.Among them,type 2 diabetes(T2DM)accounted for over 90%of the total incidence rate of diabetes,also known as adult diabetes.The high age group was 35?40 years old,and most of them had different degrees of insulin resistance.Insulin can promote the uptake and utilization of glucose in the whole body,promote the synthesis of glycogen in liver and muscle,inhibit gluconeogenesis and reduce blood glucose level.Islet ? cells are the only cells that secrete insulin.The maintenance of normal cell number and function is the necessary condition for the body to secrete enough insulin and maintain the balance of blood glucose.An early description of Potentilla discolor Bunge(PDB),also known as Azure white,pulsatilla,white leaf,and chicken leg seedlings,is contained in the "Rescue Materia Medica".P.discolor is sweet,bitter,non-toxic,and is directed to the liver,stomach,and large intestine.It clears heat,detoxifies,stops bleeding,and reduces swelling.P.discolor is used to treat a wide range of diseases,such as dysentery,malaria,lung sputum,hemoptysis,hematemesis,uterine bleeding,bloating,sores,and several other symptoms.In folk medicine,it is often used with good results for the treatment of diabetes.Experimental research shows that P.discolor has protective effect on type 2 diabetic islet? cells,and can improve insulin resistance(IR)and lipid metabolism flounder to achieve hypoglycemic effect.Metformin is a clinical first-line drug for the treatment of type 2 diabetes mellitus.It is an insulin sensitizer,which can effectively inhibit the liver glucose output,promote the liver cells,fat and muscle to absorb a large amount of glucose from the blood,with obvious hypoglycemic effect.It has been used for many years in clinical practice.Therefore,through animal experiments,this study observed and compared the therapeutic effect of water extract of Potentilla discolor combined with metformin and metformin on type 2 diabetic rats and the effect on the function of islet ? cells.Objective:The effects of aqueous extract of Potentilla discolor combined with metformin on glucose and lipid metabolism and islet function in type 2 diabetic rats were observed.Methods:A 4-week-old SPF male SD rat was administrated with a high-fat emulsion and low-dose streptozocin(STZ)to establish a type 2 diabetes model.They were divided into 7 groups:normal group,model group,positive control group(metformin hydrochloride),low-dose group and high-dose group,low-dose combined metformin group and high-dose combined metformin group.Specific technology was applied to measure serum glucose,total cholesterol(TC),high density lipoprotein(HDL),low density lipoprotein(LDL)and triglycerides(TG).ELISA was applied to measure serum insulin levels.Pancreatic histology was examined by HE staining.Immunohistochemistry(IHC)was employed to examine Forkhead box O1(FoxO1),p-FoxO1 and insulin protein expression.Results:Compared with metformin alone,the aqueous extract of Potentilla discolor combined with metformin can groups decreased random blood glucose and TC levels,increased insulin expression,promoted the release of insulin,decreased the expression of FoxO1 protein in the nucleus,induced the phosphorylation of FoxO1 protein and its getting out of the nucleus.Conclusion:In the treatment of T2DM,the water extract of Potentilla discolor combined with metformin can regulate FoxO1 and its phosphorylation,promote insulin release and improve the dysfunction of pancreatic ? cell.Compared with metformin alone,the combination of traditional Chinese and Western medicine can protect and repair the pancreatic cells of type 2 diabetic rats better and improve insulin sensitivity. |
