In recent years, Diabetes mellitus (DM) should be also pay more attention with the development of againsting serious illness study in domestic and international. According to dates provided by International Diabetes Federation, there are more than 250 million people with diabetes in global at present. If any active measures weren't taken, this number will be increased to 380 million in twenty years. The study of Diabetes mellitus is as important as cancer and cardiovascular disease in China. This topic study for a pharmacological of Potentilla discolor Bunge. in vivo and vitro based on the folk herb medicine from Potentilla Discolor Bunge used to treating DM and search special effects anti-diabetic drug in Chinese Treaditional medicine.The study methods adopted anti-free radical model in vitro and pharmacological study in vivo which is according to free radical theory aboat diabete. These courses is order to select anti-diabetes active ingredient in Potentilla discolor Bunge. Experiment in vitro revealed the activity of different extracts in Potentilla discolor Bunge to eliminate·OH and inhibit nitration reaction. Experiment in vivo used alloxan induced and copied diabetes rat model. At the same time, set medicine dose is acording to maximum tolerated dose in acute toxicity test, and positive control dose is on the basis of specification. Which is a chemical means with the different extracts from Potentilla discolor Bunge. Different Potentilla discolor Bunge extracts and positive drug were gastric perfusion to treat DM rat. The biochemical indices were detected such as glucose , total cholesterol, triglyceride, high density lipoprotein and low density lipoprotein in the end, but glucose was detected in every week in order to observe its change .Then pathological testing was observed on heart, liver, kidney and artery.The results show Potentilla discolor Bunge different extractions eliminate·OH and H2O2 and inhibit nitration ability and anti-diabetes model of nitration reaction predict that 95% ethanol was the anti-diabetic material base in vitro. This extraction eliminate rate of·OH and inhibit rate of nitration reached 26.9% and 58.6%, respectively. So, Potentilla discolor Bunge can anti-free radicals. The animal experiment shows that, the glu of treated rats have been obviously decreased which compared with the model group P<0.05 in vivo (Tab.4-4). The Total cholesterol and Low density lipoprotein were also obviously decreased which compared with the model group P<0.05(Tab.4-5 Tab.4-8). In summary, presentation a description their current trends as follows:Hypoglycemic effect (Fig.4-4):Metformin HCl> the 95% exteacts of Potentilla discolor Bunge > Tangniaole; Total cholesterol (Fig.4-5):the 95% exteacts of Potentilla discolor Bunge < Metformin HCl < Tangniaole; Low density lipoprotein (Fig.4-8):Metformin HCl~the 95% exteacts of Potentilla discolor Bunge < Tangniaole; Triglyceride (Fig.4-6):Metformin HCl< Tangniaole < the 95% exteactions of Potentilla discolor Bunge; High density lipoprotein (Fig.4-7):Tangniaole > the 95% exteacts of Potentilla discolor Bunge~Metformin HCl. The biochemical indices reveal that Potentilla discolor Bunge have the effect of hypoglycemic and lipid-lowering. Compare to two positive control drug which are known, Potentilla discolor is superior to Tangniaole, but inferior to Metformin HCl. For experiment time too short, induce that groups have no obvious differeces in pathological testing. At the same time, there are no obvious abnormal in Potentilla discolor, Bung treated group, which discripted that its Side effect isn't so heavy and it is safe.This project results show the efficiency of our studies methods by different extractions of the Potentilla discolor Bunge plant pharmacodynamics screening. Besides, its antihyperglycaemic and antihyperlipidaetic activity are belong to certain compositions in Potentilla discolor Bunge. This pharmacology study offered experiment basis and theoretical guidance for Potentilla discolor Bunge phytochemistry and searching new hypoglycemia medicine. Besides, the nitrition model was used in DM screening of Chinese herb medicine in vitro that is a innovations in this paper.
