| Objective:To investigate the characteristics of"skin microenvironment"in the guinea pig model of dermatitis eczema like lesion with spleen deficiency and dampness accumulation and the regulatory effect and mechanism of Xiaofeng powder and Sijunzi Decoction on the disorder of"skin microenvironment"in the guinea pig model of dermatitis eczema like lesion with spleen deficiency and dampness accumulation.Methods:1.Study on the skin microenvironment of guinea pig model of dermatitis eczema like lesion with spleen deficiency and dampness accumulation:24 guinea pigs were randomly divided into normal control group,dermatitis eczema group and dermatitis eczema group with spleen deficiency and dampness accumulation,8 in each group.The dermatitis eczema group was induced by 2,4-dinitrochlorobenzene(DNCB),and the dermatitis eczema group with spleen deficiency and dampness accumulation was induced by 2,4-dinitrochlorobenzene(DNCB),4-dinitrochlorobenzene(DNCB)and cold extract of senna leaf were used to establish the model.The skin moisture content,water loss and weight gain of guinea pigs were measured by mpa6/10 multifunctional skin tester(CK company,Germany)every week.The pathological changes of guinea pigs were observed after he staining.The serum Ig E,IL-4,IL-17,CD4~+and CD8~+concentrations were detected by ELISA.The expression of AQP3 protein in skin lesions was detected by Western blot The expression of AQP3 m RNA in skin lesions was detected by flow cytometry.2.Regulation experiment of Xiaofeng powder and Sijunzi Decoction on"skin microenvironment"obstacle of spleen deficiency and dampness accumulation dermatitis eczema like lesion guinea pig model:36 guinea pigs with spleen deficiency and dampness accumulation dermatitis eczema like lesion model were randomly divided into model control group,positive drug control group and Xiaofeng powder and Sijunzi Decoction group,with 12 in each group.The skin moisture content,water loss and weight gain of guinea pigs were measured by mpa6/10 multifunctional skin tester(CK company,Germany)every week.The pathological changes of skin lesions were observed after he staining.The serum Ig E,IL-4,IL-17,CD4~+and CD8~+concentrations were detected by ELISA.The expression of AQP3 protein was detected by Western blot,and the expression of AQP3 m RNA was detected by q PCR.Results:1.Compared with the normal group,the skin of the control group had erythema,a little papule,erosive exudation,part of blood scab,and frequent licking and scratching sensitization area.2.The skin of guinea pig is fluffy and lusterless,the food intake is reduced,the body moves lazily,piles up,and the stool is thin and soft.The skin moisture content decreased(P<0.01)and the water loss increased(P<0.01),the weight gain decreased(P<0.01).The pathological examination showed hyperkeratosis of skin epidermis,hyperplasia of connective tissue of granulosa layer,spinous cell layer,basal layer cells and dermis,infiltration of inflammatory cells,increase of serum Ig E,IL-17,CD8~+concentration(P<0.01),increase of IL-4,CD4~+concentration and CD4~+concentration(P<0.01)/CD8~+ratio decreased(P<0.01),and the relative expression of AQP3 in skin,lung and intestine increased(P<0.01).2.Compared with the model group,the skin lesion score of Xiaofeng powder combined with Sijunzi Decoction group was decreased(P<0.01),the skin moisture content was increased(P<0.01),the skin water dispersion loss was decreased(P<0.01),the weight gain was increased(P<0.01),and the pathological changes of skin keratinization,the proliferation of granular layer,spinous cell layer,basal layer cells,and dermis fibroconnective tissue and the infiltration of inflammatory cells were different The levels of Ig E,IL-17 and CD8~+in serum were decreased(P<0.01),the levels of IL-4 and CD4~+in serum and the ratio of CD4~+/CD8~+were increased(P<0.01),and the relative expression of AQP3 in skin and intestinal tissue was decreased(P<0.01).Conclusion:the skin water content and water loss,histopathological changes,AQP3 m RNA and protein expression of the guinea pig model of dermatitis eczema like lesions with spleen deficiency and dampness accumulation have obvious characteristics.Xiaofeng powder and Sijunzi Decoction can regulate the water regulation,immune function,AQP3 m RNA and protein expression,inhibit the pro-inflammatory factors,It can repair the barrier function of epidermis and improve the microenvironment of fur. |
