Ascites syndrome(AS)is a kind of common nutritional deficiency disease,the pathogenic factors are complex and diverse,hypoxia is the initiating factor,and pulmonary hypertension is the pathogenesis.Erythropoietin(EPO)is a hormone mainly secreted by kidney,which has the functions of promoting red blood cell production,regulating autophagy,and anti-fibrosis.During the pathogenesis of AS,EPO participates in the formation of pulmonary hypertension and affects the occurrence of AS.The purpose of this experiment was to study the mechanism of action of kidney EPO and its related factors in the formation of AS in broilers,observe the function of the compound traditional Chinese medicine selected by this research group.MethodThe AS model was constructed by the multi-factor method of adding 3% lard and 4% fish meal to basal feed and 0.12% Na Cl to drinking water at the low temperature of 9-11℃.A total of 125 1-day-old Ross broilers were adaptive feeding 8 days,then were randomly divided into normal group,AS model group,compound traditional Chinese medicine high dose group(fed with AS model group and respectively given a dose of 2 m L/kg·bw·d compound traditional Chinese medicine in drinking water),compound traditional Chinese medicine low dose group(fed with AS model group and respectively given a dose of 1m L/kg·bw·d compound traditional Chinese medicine in drinking water)and L-Arg group(fed with AS model group and added 1% L-Arg in diet).Selected 5 broilers randomly at 15,25,35 and 45 days of age,weighted,took samples of blood,heart and kidney,observed the clinical symptoms and autopsy changes,calculated the ascites heart index(AHI),HE staining was used to observe the pathological changes of kidney at the age of 35 days,RBC,PCV,Hb in blood of broilers at each day-age were detected by automatic blood cell analyzer,the expression of HIF-1α,KIM-1,EPO,EPOR,CD131,LC3-Ⅱ in kidney of broilers at each day-age were detected by ELISA.Result1.Compared with the normal group,broilers in AS model group were depression,the feathers were messy,the abdomen was swollen and had a wave feeling,a large amount of yellow liquid could be observed by puncture.Pathological necropsy showed lung and liver congestion and swelling,right ventricular hypertrophy with pericardial effusion,kidney congestion and swelling with white deposits,intestinal wall congestion or bleeding spots of varying sizes.The clinical symptoms and pathological changes of broilers in the high-and low-dose groups of compound traditional Chinese medicine and L-Arg group were all relieved.2.At 35 days of age,compared with the normal group,the kidney tissue cells of broilers in AS model group were arranged in disorder,the epithelial cells of renal tubules were exfoliated and vacuolized,the interstitials were filled with inflammatory cells;compared with AS model group,the high-and low-dose groups of compound traditional Chinese medicine and the L-Arg group were showed amelioration.3.Compared with the normal group,broilers in AS model group showed that the body weight was significantly decreased(P<0.01,P<0.05),the AHI,RBC,PCV,Hb in blood,the expression of HIF-1α,EPO in kidney were significantly increased at each day-age(P<0.01),the expression of EPOR in kidney was significantly decreased at 15 days of age(P<0.01),no significant difference of CD131 protein expression in kidney at each day-age(P>0.05),the expression of KIM-1 in kidney at 25,35 days of age and LC3-Ⅱ in kidney at 25,35,45 days of age were significantly increased(P<0.01,P<0.05).4.Compared with AS model group,broilers in high-and low-dose groups of compound traditional Chinese medicine showed that the body weight was significantly increased(P<0.01,P<0.05),the AHI,RBC,PCV,Hb in blood,the expression of HIF-1α,EPO in kidney were significantly decreased at each day-age(P<0.01),the expression of EPO in kidney was significantly increased in high dose groups of compound traditional Chinese medicine at 35,45 days of age(P<0.01,P<0.05),the expression of EPO in kidney was significantly decreased in low dose groups of compound traditional Chinese medicine at 25,35 days of age(P<0.05),the expression of EPOR in kidney was significantly increased in high-and low-dose groups of compound traditional Chinese medicine at 15 days of age(P<0.05),no significant difference of CD131 protein expression in kidney in each group at each day-age(P>0.05),the expression of KIM-1 and LC3-Ⅱ in kidney was significantly decreased in high-and low-dose groups of compound traditional Chinese medicine at 25,35,45 days of age(P<0.01,P<0.05).5.Compared with AS model group,broilers in L-Arg group showed that the body weight was significantly increased(P<0.05),the AHI,RBC,PCV,Hb in blood,the expression of HIF-1α in kidney were significantly decreased at each day-age(P<0.01,P<0.05),the expression of EPO in kidney at 25,35 days of age and KIM-1,LC3-Ⅱ in kidney at 25,35,45 days of age were significantly decreased(P<0.01,P<0.05),no significant difference of EPOR and CD131 protein expression in kidney at each day-age(P>0.05).Conclusion1.During the occurrence and development of ascites syndrome in broilers,EPO production in kidney was increased,a large number of red blood cells were produced that made the blood sticky,which was involved in the occurrence and development of ascites syndrome in broilers,meanwhile,the reduce of renal erythropoietin receptor expression,renal hypoxia injury and the increase of autophagy were also involved in the occurrence and development of ascites syndrome in broilers.2.The compound traditional Chinese medicine was made up of astragalus membranaceus,poria cocos,angelica sinensis,pericarpium arecae,salvia miltiorrhiza,rhizoma chuanxiong,liquorice and so on has the function of replenishing qi and blood,invigorating qi and promoting blood circulation,clearing damp and promoting diuresis,it can effectively prevent the occurrence and development of ascites syndrome in broilers by regulating the function of renal erythropoietin and its receptor,regulating blood erythropoiesis,improving blood consistency,improving renal hypoxia and autophagy.3.L-Arg can effectively prevent the occurrence and development of ascites syndrome in broilers by regulating renal erythropoietin,regulating blood erythropoiesis,improving blood consistency,improving renal hypoxia and autophagy injury.
