| Ascites Syndrome is a group of serious metabolic diseases with serious causes and high incidence.Studies have confirmed that traditional Chinese medicine has the characteristics of low residue,low toxicity and side effects and can effectively prevent and treat AS.Studies have shown that hepatic and renal fibrosis caused by hypoxia is closely related to the formation of ascites.The objective of the research was to investigate the mechanism of compound traditional Chinese medicine on hepatic and renal fibrosis in broiler ascites syndrome.Method A total of 309 7-day-old Ross broilers were randomly divided into five groups,the group None was bred with basal feed,and the group Control was bred with mixed feed(9-11 ℃,added 3 % lard and 4 % fish meal to basal feed,added 0.12% Na Cl to drinking water),compound traditional Chinese medicine group(High,Low)was given 2,0.5g·(kg·d)-1 of the CTCM in drinking water respectively on this basis,Group L-Arginine(with 1% L-arginine in diet).Selected 10 broilers randomly at 15,25,35 and 45 days of age,weighted and killed to observe the pathological change of broilers,and severed heart to measure ascites heart index,HE and Masson staining was used to observe the pathological changes of hepatic and renal and the changes of collagen fiber content at the age of 35 days,and Using ELISA to detect the content of HIF-1α,NOX4,Ang II,TGF-β1 in hepatic and renal tissue.Result 1、Broilers in group Control were depression,comb and beard cyanosis with undulating abdominal distention,light yellow or straw yellow ascites could be observed by puncture,pericardial effusion could be observed by autopsy,hypertrophy of heart,,and hyperemia and swelling of hepatic,the attachment of gelatinous clumps,and hyperemia and swelling of renal.The above symptoms could be improved to varying degrees in each dose of CTCM groups and L-Arg group.2、Compared with group None,the ascites heart index of AS in each age of group Control was significantly increased(P<0.01),body weight significantly decreased(P<0.01 or P<0.05).Compared with the group Control,body weight of each age of high doze of compound traditional Chinese medicine group,35 days of low doze of compound traditional Chinese medicine group and 15,25,35 days of L-Arg group significantly increased(P<0.01 or P<0.05).The ascites heart index of AS in the compound traditional Chinese medicine groups and the L-Arg group were significantly decreased(P<0.01 or P<0.05).3、At 35 days of age,compared with group None,hepatic and renal tissue in group Control were lose normal structure,with irregular cell alignment,inflammatory cell infiltration,collagen fibril increased;Compared with group Control,the compound traditional Chinese medicine groups and the L-Arg group were showed amelioration of different degree.4、At 35 days of age,compared with group None,protein expression levels of HIF-1α,NOX4,Ang II,TGF-β1 in hepatic and renal of broilers in group Control were increased significantly(P<0.01 or P<0.05).Compared with group Control,the above indexes were improved in each dose of traditional Chinese medicine groups and L-Arg group(P<0.01 or P<0.05).Conclusion 1、AHI and the expression levels of HIF-1α,NOX4,Ang II,TGF-β1 at 35 days of age in hepatic and renal tissues of AS were increased,with low body weight,HE and Masson staining showed fibrosis in hepatic and renal tissues.Indicating that HIF-1α through regulating NOX4 and Ang II increased oxidative stress and inflammation,activate the TGF-β1,and participate in the pathological process of hepatic fibrosis and promoties the progression of AS.2、The Compound traditional Chinese medicine with functions of invigorating qi and blood,activating blood and qi,clearing damp and promoting diuresis and L-arginine can augmented body weight,down-regulate the AHI and the protein expression levels of HIF-1α,NOX4,Ang II and TGF-β1 protein at 35 days of age in hepatic and renal in AS.Indicating that the compound traditional Chinese medicine and L-arginine can Inhibit oxidative stress and inflammation,inhibits the pathological development of hepatic and renal fibrosis and effectively prevent and treat AS. |
