Objective:Broiler ascites syndrome is a fatal nutritional metabolic disease, its pathogenesis is complex, the cause is various, the mortality rate is high.There is no effective control measures for this diease,causing a huge economic losses for commercial broiler breeding in the worldwide.And pulmonary arterial hypertension has been shown as the key step in broiler ascites syndrome.Vascular lesions is the initiation factor for the study of pulmonary arterial hypertension.lt is of great significance to explore the pathogenesis of pulmonary hypertension and broiler ascites syndrome from the perspective of vascular physiology and pathology. By detecting gene and protein content of related factors myocardin SM22 alpha and KLF-4 in smooth muscle phenotypic transformation, research the compound Chinese medicine and phenotype transformation of smooth muscle in broiler ascites syndrome in which pulmonary hypertension plays a huge roleIn this study, we successfully established broiler ascites syndrome model by the multi factor method. 330 broilers age of 8, were randomly divided into 6 groups:blank group, the AS model group, the compound Chinese medicine high dose group (2 mL/kg'd), the compound Chinese medicine agent group (1mL/kg·d), the compound Chinese medicine low dose group (0.5 mL/kg·d) and positive drug group (1%L-Arg). In 15,25,35,45 days of age,selected five broilers in each group randomly, pathological anatomy and take heart and lung tissue, detect Ascites heart index (Ascites heart index, AHI)The morphology of pulmonary artery was observated by trichromatic staining method (massion). We determinate the mRNA and protein levels of myocardin, SM22 alpha and klf-4 in pulmonary vascular by fluorescence quantitative PCR and ELISA in various day-old. To study resistance mechanism of traditional Chinese medicine to broiler ascites and the mechanism of smooth muscle pathological changes in broiler ascites syndrome. The results are as follows:1. Compared with the blank group,15,25,35,45 days of age model group pulmonary vascular membrane swelling, muscle fiber fracture, clutter to the endometrium. Compared with model group, the compound Chinese medicine high dose group, the compound Chinese medicine middle-dose group, compound traditional Chinese medicine (TCM) low dose group and positive group on a different day age there are different levels of repair endothelial and smooth muscle function. That the smooth muscle cells have lesions in broiler ascites syndrome,compound traditional Chinese medicine has an important role in inhibiting smooth muscle lesions.2.Fluorescence quantitative PCR results showed that compared with the blank group, model group,15?35 and 45 days of age pulmonary vascular KLF-4 gene significantly increased (P< 0.05),15,25,35 and 45 days of age pulmonary vascular SM22 alpha and Myocardin gene significantly reduced (P< 0.05); Compared with the model group, high dose group 15,35, and 45 days of age pulmonary vascular KLF-4 gene decreased significantly (P< 0.05),15,25,35 and 45 days of age pulmonary vascular SM22 alpha and myocardin gene was significantly increased (P< 0.05), Middle compound Chinese medicine dose group of 15 and 35 days of age pulmonary vascular KLF-4 significantly reduced (P< 0.05),35 and 45 days of age pulmonary vascular Myocardin gene significantly increased (P< 0.05),15,25,35 and 45 days of age pulmonary vascular SM22 alpha gene significantly higher (P< 0.05);Low compound chinese medicine dose group of pulmonary vascular KLF-4 day age of 35 significantly lower (P< 0.05),25 and 35 days of age pulmonary vascular SM22 alpha gene significantly higher (P< 0.05);Positive group of 35 and 45 days of age 15, pulmonary vascular KLF-4 gene significantly reduced (P< 0.05),15,35 and 45 days of age pulmonary vascular Myocardin gene significantly higher (P< 0.05),15,25, 35 and 45 days of age pulmonary vascular SM22 alpha gene significantly increased (P< 0.05), 3.Elisa results show that Compared to blank group,the protein expression amount in model group KLF-4(15,25, and 35 days of age) was significantly increased (P< 0.05), SM22 alpha (15,25,35 and 45 days of age) protein decreased significantly (P< 0.05), myocardin(15,25,35 and 45 days of age) protein significantly reduced (P< 0.05).Compared with model group, the compound Chinese medicine high dose group of 25 and 35 days of age pulmonary vascular KLF-4 protein decreased significantly (P< 0.05), the age of 15 and 25 arteries SM22 alpha protein increased significantly (P< 0.05),15,25 and 35 days of age arteries Myocardin protein increased significantly (P< 0.05);Compound Chinese medicine middle dose group of 15 days of age pulmonary vascular KLF-4 proteindecreased significantly (P< 0.05),15,25 and 35 days of age pulmonary vascular Myocardin significantly increased (P< 0.05),15,25,35 and 45 days of age pulmonary vascular SM22 alpha protein increased significantly (P< 0.05); Compound traditional Chinese medicine low dose group of 15 and 25 days of age pulmonary vascular Myocardin protein increased significantly (P< 0.05),15,25 and 45 days of age pulmonary vascular SM22 alpha protein increased significantly (P< 0.05);Positive group of 15,25 and 35 days of age pulmonary vascular KLF-4 protein decreased significantly (P < 0.05),15,25 and 35 days of age pulmonary vascular Myocardin and SM22 alpha protein increased significantly (P< 0.05)Conclusion:1.Vascular smooth muscle phenotype transformation factor myocardin?KLF-4 and sm22 a involved in the pathological development of broiler ascites syndrome.2.The compound Chinese medicine can inhibit the artery vascular smooth muscle phenotype transformation to prevention and control of broiler ascites syndrome effectively.3. L-Arg can inhibit artery vascular smooth muscle phenotype transformation to prevention and control of broiler ascites syndrome effectively.
