Effect Of The Compound Recipe Of Traditional Chinese Medicine On Liver Fibrosis Of Broilers With Ascites Syndrom

Ascites Syndrome(AS)is a kind of commom metabolic with complicated etiological factors,which keeps high incidence and unfavourable prognosis.As natural productions,demonstrated to prevent and treat AS effectively,traditional Chinese drugs have low residue and adverse effects.The pathological changes of liver of broilers in the progress of AS are correlated with fibrosis factors like TGF-β1,and TGF-β1/Smad pathway closely.The objective of the research was to investigate the changes of collagens,fibrosis factors and TGF-β1/Smad pathway of livers in broilers with AS,and the mechanism of treatment and prevention of the compound recipe of traditional Chinese medicine on AS.MethodA total of 309 7-day-old Ross broilers randomly divided into 6 groups,Group None(N,breed with standard diet at room temperature),and others induced AS with multiple factos(high-energy diet derived from standard diet added with 3%lard and 4%fish meal and 0.12%Na+ in drinking water at 9-11 ℃),including Group Control(C,given mo medicine),Group L-Arginine(L,with 1%L-arginine in diet)and Compound recipe of the traditional Chinese medicine Groups(T1,T2,T3,respectively given a dose of 2,1,0.5 g/kg·d of compound recipe of the traditional Chinese medicine in drinking water).Selected 10 broilers randomly at 15,25,35 and 45 days of age,weighted and killed to observe the pathological change of broilers,and severed heart,lung,liver and spleen to measure viscera indexes,and studied the histopathology changes of liver of broilers,and determine the levels of α-SMA、collagen Ⅰ,collagen Ⅲ,HIF-1α,TGF-β1,TβRI and CTGF in liver by ELISA and the relative expressions of HIF-1α、TGF-β1、TβRI、CTGF、Smad2、Smad3、Smad7 mRNA in liver by QT-PCR.Result1 Big-bellied broilers in Group C were in low spirits and weight,with strew liquid in abdominal cavity.Autopsy results indicated hydropericardium,hypertrophy of right heart,pulmonary ecchymosis and hepatic ecchymosis.All of these symptoms could be improved by compound recipe of the traditional Chinese medicine and L-arginine.2 At 15,25,35 and 45 days of age,compared with Group N,the incidence of AS,ascites heart index,heart index,lung index and liver index increased significantly in Group C(P<0.01 or P<0.05),and body weight and spleen index significantly decreased(P<0.01 or P<0.05)in Group C.Compared with Group C,compound recipe of the traditional Chinese medicine and L-arginine improved all items above(P<0.01 or P<0.05).3 At 15,25,35 and 45 days of age,compared with Group N,the level of a-SMA、collagen I,collagenⅢ,HIF-1α,TGF-β1,TβRI and CTGF in liver significantly increased(P<0.01 or P<0.05)in Group C.Compared with Group C,compound recipe of the traditional Chinese medicine and L-arginine significantly decreased the level of α-SMA、collagen Ⅰ,collagen Ⅲ,HIF-1α,TGF-β1,TβRI and CTGF in liver(P<0.01 or P<0.05).4 At 15,25,35 and 45 days of age,compared with Group N,the relative expression of HIF-1α、TGF-β1、TβRI、CTGF、Smad2 and Smad3 mRNA significantly increased(P<0.01 or P<0.05),and the relative expression of Smad7 mRNA significantly decreased(P<0.01 or P<0.05)in Group C.Compared with Group C,the relative expression of HIF-1α、TGF-β1、TβRI、CTGF、Smad2 and Smad3 mRNA significantly decreased(P<0.01 or P<0.05),and the relative expression of Smad7 mRNA significantly increased(P<0.01 or P<0.05)in Group Tl,T2,T3 and L.Conclusion1 In liver of broilers with AS,the AHI,heart index,lung index and liver index,and the level ofα-SMA、collagen Ⅰ,collagen Ⅲ,HIF-1α,TGF-β1,TβRI and CTGF,and the relative expression of HIF-1α、TGF-/β1、TβRI、CTGF、Smad2 and Smad3 mRNA increased,with low body weight,spleen index and the relative expression of Smad7 mRNA.AS regulates fibrosis factors via TGF/Smad pathway to promote the liver fibrosis,participating the development of AS.2 The compound recipe of the traditional Chinese medicine and L-arginine controlled the AHI,heart index,lung index and liver index,and the level of α-SMA、collagen Ⅰ,collagen Ⅲ,HIF-1α,TGF-β1,TβRI and CTGF,and the relative expression of HIF-1α、TGF-/β1、TβRI、CTGF、Smad2 and Smad3 mRNA,and augmented body weight,spleen index and the relative expression of Smad7 mRNA in liver of broilers with AS,to prevent and treat AS by inhibiting the level of fibrosis factors via TGF/Smad pathway.
