In this thesis,the Panax notoginseng saponin extract was used as the research object to construct the saponins chemical substance library.The complex chemical composition of Panax notoginseng is the basis for its good efficacy.At present,more than 200 kinds of compounds have been discovered from Sanqi,including saponins,flavonoids and cyclic peptides.Panax notoginseng compounds are the main active constituents in Panax notoginseng extract.There are many kinds of notoginsenoside compounds and complex structure.And the high-purity standard control is difficult to obtain,so that it is difficult to directly measure all the saponin compounds.Therefore,the high-performance liquid chromatography is usually used to determine notoginsenoside R1,ginsenoside Rg1,and ginsenoside Rd.Conduct quality evaluation.However,due to the complex chemical composition and variety of notoginsenosides,the results measured by this method are not necessarily accurate,and it is easy to fake adulteration.Therefore,it is extremely important to construct a reservoir of notoginsenosides.The main research contents and results include the following.Several parts:Thin-Layer Chromatography(TLC)and High Performance Liquid Chromatography(HPLC)methods for the determination of notoginsenosides were determined.Under this test method,all the total saponins of Panax notoginseng can be detected.A compound with high resolution.Based on the analytical method,the extract of Panax notoginseng saponins was separated and purified,and separated by silica gel and dextran gel column chromatography to obtain 11 chemical components of notoginsenosides;component B-10 i The component B-10 f and the component B-10 h were further purified by Super30 and C18 preparative liquid phase to obtain three notoginsenoside monomer compounds,and three Sanqi were identified by physical and chemical properties,spectral information,chromatographic information and NMR information.Saponin monomer11 compounds of notoginsenosides: 52.5 g of B-10 a component,mainly pigments,no target compound;38 g of B-10 b component,mainly compounds of No.1 and No.2;component of B-10 c 246 g,mainly compound No.2;B-10 d component 60 g,mainly compounds No.4 and No.5;B-10 e component 259 g,mainly No.4,No.5,No.6,No.7,and No.8 Compound;B-10 f component 64 g,mainly 9 and 10 compounds;B-10 g component 253 g,mainly 12,13compound;B-10 g component 31 g,mainly 14 th,15 Compound No.;B-10 h component 52 g,mainly complex components,but most of them are target compounds;B-10 i component 19 g,mainly complex components,but less target compound;B-10 j component 100 g,For the pure methanol elution component,the total mass yield was 85.7%,and the chromatographic information of each component was obtained by high performance liquid chromatography analysis to establish a chemical constituents library of notoginsenosides;3 notoginsenoside monomer compounds: compound No.1 is notoginsenoside R1,mass700 mg,purity greater than 99.2%;compound No.2 is ginsenoside Rg1,mass 200 g,purity greater than 98.8%;compound 11 is ginseng Saponin Rd,mass 35 g,purity 98.2%.Finally,based on 11 chemical components of notoginsenoside and three notoginsenoside monomer compounds,the reservoir of notoginsenosides was constructed to establish the basis for the subsequent quality evaluation and activity study of notoginsenosides.
